Long Non-Coding RNAs as Molecular Biomarkers in Cholangiocarcinoma

Abstract: Cholangiocarcinoma (CCA) is a biliary system cancer that has the characteristics of strong invasiveness, poor prognosis, and few therapy choices. Furthermore, the absence of precise biomarkers for early identification and prognosis makes it hard to intervene in the early phase of initial diagnosis or recurring cholangiocarcinoma following surgery. Encouragingly, previous studies found that long non-coding RNA (lncRNA), a subgroup of RNA that is more than 200 nucleotides long, can affect cell proliferation, mig… Show more

“…Moreover, long non-coding RNAs (lncRNAs) are characterized as non-coding RNAs whose transcripts are longer than 200 nucleotides lacking the ability to code for proteins but influencing tumorigenesis, which are also implicated in the molecular biomarkers of CCA ( 136 , 137 ). For instance, lncRNA PCAT1 was remarkably upregulated in both eCCA tissues and cell lines ( 109 ).…”

“…Like miRNA, lncRNA also play dual role, as tumor suppressor role or oncogenic role. Some specific lncRNAs that have been studied in relation to Cholangiocarcinoma, such as Prostate cancer-associated transcript 1(PCAT1), Neighboring enhancer of FOXA2 (NEF), Metastasis-associated lung adenocarcinoma transcript 1(MALAT1), H19, Colon cancer-associated transcript 1 (CCAT1), and ZEB1 Antisense RNA 1(ZEB-AS1) ( Yang et al ., 2020 ; Wu et al ., 2022 ). Exostosin1 (EXT1): EXT1 is a gene encodes an endoplasmic reticulum-resident type II transmembrane glycosyltransferase involved in the chain elongation step of heparan sulfate biosynthesis.…”

“…As an important regulatory factor in the human genome, lncRNA regulates histone modification, DNA methylation, or chromosome reconstruction through epigenetic regulation, transcriptional or post-transcriptional regulation, and other mechanisms to silence or activate genes, and then dynamically control disease-related gene changes and some important biological processes [ 4 ]. The abnormal expression of lncRNA can participate in both tumor inhibition and carcinogenesis [ 5 ]. Currently, many lncRNAs have been confirmed to be abnormally expressed in various organ tumors.…”
