The patterns of reactivity of antisera raised in rabbits to human myelin and to the basic protein of human myelin were studied by immunofluorescence, using frozen sections from autologous as well as heterologous central nervous system (CNS) and smears of myelin and a synaptosomal membrane fraction of CNS. Whole myelin induced antibodies which reacted with myelinated areas in frozen sections of the CNS in a homogeneous pattern, and with smears of myelin and the synaptosomal membrane fraction; this activity was absorbed from serum by myelin, by the water-insoluble fraction of myelin, but not by the basic protein of myelin. The basic protein of myelin induced antibodies which reacted with myelinated areas in the CNS in a finely speckled pattern, and with smears of myelin and the synaptosomal membrane fraction; this activity was absorbed from serum by myelin, by myelin basic protein and partly by histones. This evidence of cross-reactivity in immunofluorescence tests between the myelin basic protein and histones was corroborated by partial competitive inhibition between histone and the myelin basic protein in the radioimmunoassay for antibody to basic protein. An antigen similar to myelin basic protein is present in the synaptosomal membrane fraction of CNS and hence synaptosomes are potential reactive sites for the pathogenic immune response in experimental allergic encephalomyelitis.

Beutner et al (16) reported similar findings in rabbits injected with native thyroid extract incorporated into complete Freund's adjuvant. As in the autoimmune phenomenon produced by injection of native thyroglobulin incorporated into complete Freund's adjuvant (17), the data in the present study show no correlation between the level of circulating antithyroglobulin and severity of lesions.…”

Experimental autoimmunity was produced in rabbits following injection of altered homologous thyroglobulin. The thyroglobulin was altered by coupling to chemically defined haptens and by heating. With some preparations antibody to native thyroglobulin as well as thyroid lesions were produced. Injections of thyroglobulin coupled to the diazonium derivatives of arsanilic acid and sulfanilic acid were effective when given in either soluble form or incorporated into incomplete Freund's adjuvant) while injections of the same preparations precipitated by alum had relatively little effect on production of antibody or induction of lesions. The injection of native thyroglobulin in soluble form, incorporated into incomplete adjuvant or precipitated by alum usually resulted in production of little or no antibody and only rarely in the formation of lesions. The injection of a heterologous thyroglobulin into rabbits resulted in the production of antibody reacting with both the heterologous and rabbit thyroglobulin, but no thyroid lesions were observed.

A. Fluorescent Antibody Labeling Compounds which emit light in one wave length range when illuminated by light of a shorter wave length range are fluorescent. Most fluorescent compounds that have been studied are excited by light in the ultraviolet or near ultraviolet region and emit light in the visible range. Fluorescent dyes are called fluorochromes. The coupling of fluorochromes to antibodies forms the basis of staining with labeled antibody. Briefly, three methods of coupling fluorescent dyes to proteins have been utilized to prepare conjugates for IF staining. (As used here the term "conjugates" refers to fluorochrome-labeled proteins.) According to the original procedure of Coons, Creech, Jones, and Berliner (30) an isocyanate derivative of fluorescein is prepared by reaction of amino group of the fluor with phosgene.
