SUMMARY1. The rate of acetylcholine (ACh)-induced fluid secretion was measured from the main excretory duct of rabbit lacrimal glands perfused in vivo with Krebs Henseleit bicarbonate solutions.2. Perfusion with ouabain (10-5 M) decreased the rate of lacrimal gland fluid secretion to 23 % of the control value.3. Perfusion with furosemide (10-4 and 1O-3 M), which has been shown to inhibit the coupled transport of Na+ and Cl-, reversibly decreased the rate of secretion to 43 and 33 % of the control value respectively. 6. It is concluded that ACh-induced secretion of electrolytes and water is dependent upon (Na+ + K+)-activated ATPase. In addition, coupled transport of Na+ and Cl-appears to be involved in secretion.7. Basolateral location of the (Na+ + K+)-activated ATPase implies that it plays an indirect role in electrolyte and water secretion. A possible role may be to energize a secondary active transport of Cl-that is mediated by a NaCl cotransport system.

Intracellular K+ activity (aKi) and transmembrane potential differences were simultaneously measured with double-barreled K+ selective microelectrodes in superfused canine submandibular glands at rest and during stimulation. Also intracellular K+ and Na+ concentrations in the same gland were determined by chemical analyses for comparison with the intracellular K+ activity data. The activity coefficient for intracellular K+ thus obtained was 0.67, indicating that virtually all K+ ions in the cells are in diffusible and osmotically active form. Under control conditions, measured values of the resting membrane potential (Em), aK and K+ equilibrium potential (EK) were -40.2+0.6 mV (S.E., n=163), -88.5+1.4 mM (S.E., n=163), -83 mV respectively. Electrochemical potential differences for K+ across the basolateral membrane (d PKIF) were approximately ±43 mV. Acetylcholine (ACh) induced an abrupt membrane hyperpolarization followed by a temporary fall of aKi. The maximum rate of aKi change in the cytoplasm during stimulated conditions was -35.5+0.9 mM f min on the average (n= l0). These results suggest that the membrane permeability increases to K~ upon stimulation. Possible changes in membrane permeabilities to Na+ and Cl-were also discussed. Applying a microelectrode technique, LUNDBERG (1955) first recorded the resting membrane potential of cat submandibular gland cells. He found that the transmembrane potential rapidly hyperpolarized when chorda tympani was stimulated. BURGEN (1956) demonstrated that the salivary gland in the dog lost K+ to blood and saliva during the first period of secretion (K+ transient). Since then, the roles of K+ and other ions in secretory mechanisms in the gland cells have became of great concern among investigators. Intercellular material in the basal and lateral folds of parotid serous cells in four species of bats

Background: Basal folds are slender plications at the basal surface of acinar cells in the salivary glands of many mammalian species. These largely organelle-free folds increase the surface area of the basal plasmalemma manyfold and are unquestionably involved in the translocation of organic and inorganic molecules and water into the acinar cells.Methods: Specimens of salivary glands were obtained from over 230 species of live-trapped bats from major areas of the globe. Tissues for electron microscopy were fixed and processed by conventional means.Results: A number of the bat species examined had dense material in the intercellular spaces between basal and lateral folds of serous cells in the parotid gland. This intercellular material was particularly prominent in three species of New World bats, viz., Pteronotus parnellii, P. quadridens, and Phyllostomus latifolius, and in one species of Old World bats, Chalinolobus argentatus. This dense material, which has a farinaceous texture, appears not to pass through tight junctions, so it is excluded from the lumina of intercellular canaliculi and acini. The dense material originates in the acinar cells-it is carried to the membranes of the folds via coated vesicles, which empty their dense content by exocytosis into the intercellular space. Similar dense material is present in the intercellular spaces of the basal labyrinth of striated ducts in the two species of Pteronotus. The manner in which this material accumulates in the striated duct is unclear.Conclusions: Although the function of the intracellular dense material is undetermined, it appears to be placed strategically to influence molecular traffic into acinar cells or to modulate the paracellular pathway. From a comparative evolutionary perspective, we hypothesize that, in bats, the combination of basal folds and extracellular densities is associated with insectivory. Similar morphologies appear to be lacking in frugivorous or nectarivorous species. Anat.
