Localization of Paramyosin, Myosin, and a Heat Shock Protein 70 in Larval and Adult Brugia malayi

Abstract: Recombinant filarial proteins are of interest as potentially protective immunogens for lymphatic filariasis. We have previously identified paramyosin, myosin, and a heat shock protein 70 (HSP) 70 as prominent immunogens in individuals residing in an area endemic for lymphatic filariasis. Our goal in the present work was to identify the Brugia malayi tissues that contain these proteins. Polyclonal rabbit antisera with high levels of immunoglobulins to each of these proteins were prepared for use in indirect imm… Show more

“…These findings seem similar to those for HSP70 of Brugia malayi, being present in the somatic musculature, hypodermis and reproductive structures (Schmitz et al 1996) and also support the recent finding that HSP70 is one of the most abundant proteins in S. mansoni egg secretome (Cass et al 2007). “…A study of Brugia malayi has detected HSP70 in musculature, hypodermis and reproductive organs (Schmitz et al 1996), and a recent report described it as one of the most abundant surface antigens in Schistosoma mansoni egg secretions based on proteomic analysis (Cass et al 2007). “…Paramyosin (PMY), commonly known as a muscle protein of invertebrates including D. viviparus , has been considered as a promising vaccine candidate as it is transcribed throughout all developmental stages of bovine lungworms . PMY of D. viviparus displays immunomodulatory functions by binding to collagen and IgG , similar to those described for Taenia crassiceps ‐PMY .…”

“…Paramyosin has been characterized as a myofibrillar protein found only in invertebrates. In B. malayi, it is located within the longitudinal somatic musculature of L 3 , L 4 , and adult worms (21). Here we show that it is expressed in Mf as well.…”
