Localization of Fluorescein-Labeled Antibodies of Epidermal Proteins in Normal and Malignant Squamous Epithelium2

Summary.-Fluorescence microscopy was used to demonstrate differences in the urea-extractable antigens of mouse epidermis and squamous cell carcinoma. When serum-and normal tissue sediment-absorbed antisera prepared against mouse epidermal urea-extracted proteins were further absorbed with carcinoma urea antigens, antisera specific for epidermis resulted. When antisera raised against the urea-extractable proteins of mouse squamous cell carcinomata were serumand normal tissue sediment-absorbed and then further absorbed with epidermal urea antigens, antisera were prepared which stained papilloma and carcinoma, but not epidermis, and thus these antisera were not specific for carcinoma. Antisera prepared against the urea-extractable proteins of human epidermis reacted in immunodiffusion in agar with the epidermal urea proteins, but not with human squamous cell carcinoma urea proteins. Also antisera prepared against the carcinoma urea-extractable proteins reacted with these proteins in agar, but no reaction occurred with the epidermal urea-extractable proteins.

Differences in the Urea-Extracted or Fibrous Proteins of Mouse Epidermis at Various Stages of Malignant Transformation

The urea-extractable proteins of normal and hyperplastic mouse epidermis, papilloma, and squamous-cell carcinoma were fractionated by gel filtration on Sepharose 4B to yield proteins which were excluded and retarded. The level of the proteins which were excluded decreased from normal epidermis through hyperplastic epidermis, papilloma, and carcinoma, whereas the retarded protein fractions increased proportionately. Ultra-centrifugation of these urea proteins resulted in gel and supernatant fractions, each of which accounted for approximately 50% of the total proteins for normal epidermis, papilloma, and carcinoma. The gel and supernatant fractions and the proteins which were excluded and retarded by Sepharose 4B were employed in immunodiffusion in agar against antisera or IgG fractions of antisera raised against the various urea proteins. These results showed that antigenic differences existed between the fibrous proteins of normal epidermis and papilloma or carcinoma, but the latter two tissues were antigenically similar with respect to these proteins.

Urea-extractable Antigens in Normal, Benign, and Neoplastic Mouse Epidermis

Antisera were prepared in rabbits against urea-extractable proteins of normal mouse epidermis, papillomas, and squamous-cell carcinomas. The relationship of these antisera and the antigens of the three tissues was studied by immunodiffusion in agar before and after inhibition treatment of these antisera with the three antigens. These procedures revealed that (1) the epidermis has urea-extractable antigens which are present in much smaller amounts in the urea extracts of papillomas and squamous-cell carcinomas, and (2) papillomas and carcinomas have several urea-extractable antigens in common, and both the benign and malignant growths each have at least one antigen present in greater amount in one tissue than in the other. The papilloma and carcinoma antigens are present, but in small amounts in normal epidermis.
