Localization of endothelin-A and -B receptors during the postnatal development of rat cerebellum

Abstract: Intense expression of mRNA of endothelin-B receptor (ETBR) has been detected in the Bergmann glia of cerebellum by in situ hybridization, but the intracellular localization has not been reported because of the absence of a useful antibody for immunohistochemical investigations. We made polyclonal antibodies against the carboxyl terminus of human ETBR (420-442) and ETAR (403-427), and performed light- and electron-microscopic immunohistochemistry of the wild-type and ETBR-deficient (sl/sl) rat cerebella. Locali… Show more

“…It is often the case that a small difference of antibody epitope causes a difference in immunohistochemistry. We experienced cell specific localization of endothelin B receptor (ET B R) immunoreactivity by the different ET B R antibodies against synthetic peptides for the carboxyl terminus of ET B R (Furuya et al 2001). A small difference of peptide sequence produced different ET B R immunoreactivity in the endothelial cells and glial cells.…”

“…Therefore, it is necessary to view a large area to search various parts of the duct system. In the brain, we usually make 60-to 70-碌m sections by vibratome for EM immunohistochemistry (Furuya et al 2000(Furuya et al , 2001. However, pancreas is too soft to make sections by vibratome.…”

“…Rat small intestines Wild-type AR rats and Wistar rats were perfused with 4% paraformaldehyde/0.2% picric acid in 0.1 M phosphate buffer pH 7.4, and processed for light microscopic immunohistochemistry as previously described (Furuya et al, 2001). 10 渭m thick cryosections were incubated with methanol containing 0.3% H 2 O 2 for 30 minutes to inhibit endogenous peroxidase activity, and incubated with 10% NGS in PBS for 30 minutes.…”
