Localization of Carbonic Anhydrase in Living Osteoclasts with Bodipy 558/568-modified Acetazolamide, a Thiadiazole Carbonic Anhydrase Inhibitor

Abstract: SUMMARYWe describe the synthesis of Bodipy 558/568-modified acetazolamide, a fluorescent inhibitor of carbonic anhydrase and its use to localize the enzyme in living cells. The modified acetazolamide, with its specific sulfonamide group intact, labeled cells at concentrations as low as 10 -9 M, with a minimal loading time of 5 min. The staining was decreased by 57.4% by preincubating cells with unaltered acetazolamide (1:100) or with trifluoromethane sulfonamide, 6-ethoxyzolamide, and 5-(3-hydroxybenzoyl)-thio… Show more

“…Interestingly, the combination of oligothiophenes with Bodipy as an acceptor has not been reported so far. Indeed, to the best of our knowledge, the most similar system reported to date in the literature incorporated a thiophenesulfonamide and a Bodipy-modified fluorescent inhibitor that was used for cell labeling and enzyme localization …”

“…The presence of acid pH in the area of resorption is fundamental for the action of most resorptive enzymes (1, 11, 12), and decrease of the pH to nearly 4.7 at the resorption lacuna is related to the presence of hydrogen ions. Formation of hydrogen ions requires hydration of carbon dioxide (1, 11, 12), a reaction that is catalyzed by the carbonic anhydrase enzyme (13, 14). Therefore, the absence or inhibition of this enzyme alters the cycle of resorption, thereby limiting resorption (12).…”

“…A presença desses íons faz com que haja a queda do pH na área, acidificando-a 22,93,105,130 . A reação química para a formação de íons hidrogênios é catalisada por uma enzima denominada anidrase carbônica 3, 35,50,81 . Esta enzima está presente no citoplasma dos clastos 35,81 Como a reabsorção por substituição caracteriza-se pela incorporação da raiz dentária na remodelação óssea, após a constituição das lacunas de reabsorção, há a formação de tecido ósseo neste local 62,133 .…”
