Localization of binding sites for carboxyl terminal specific anti-rhodopsin monoclonal antibodies using synthetic peptides

Abstract: The binding sites for four monoclonal antibodies, rho 1D4, rho 3C2, rho 3A6, and rho 1C5, have been localized within the C-terminal region of bovine rhodopsin: Asp18'-Glu-Ala16'-Ser-Thr-Thr-Val12'-Ser-Lys-Thr-Gl u8'-Thr-Ser-Gln-Val4'-Ala-Pr o -Ala1'. Antibody binding sites were localized by using synthetic C-terminal peptides in conjunction with solid-phase competitive inhibition assays and limited proteolytic digestion of rhodopsin in conjunction with electrophoretic immunoblotting techniques. Binding of the … Show more

“…PP-1 was expressed with an N-terminal tag (ID4) that encoded part of rhodopsin (30). Briefly, the EcoRI-XhoI fragment of rabbit PP-1C␣ cDNA was subcloned into a shuttle vector plasmid pBluescript SK (Stratagene).…”

“…To improve the efficiency of purification, we used an epitope tag that has been successfully used to purify a variety of transmembrane proteins, the 1D4 epitope from the C terminus of the G protein-coupled receptor rhodopsin (23,24). The nonapeptide epitope tag incorporated into the C terminus of TRPV1 allowed us to purify TRPV1 in a single step using an immobilized monoclonal antibody, 1-D4 (Fig.…”

“…The endocytic turnover of Rh1 can be followed using an inducible and uniquely tagged Rh1. Rh1-1D4 transgenic flies express a heat-shock inducible Drosophila Rh1 carrying a specific tag, 1D4, derived from the C terminus of bovine Rh1 (19). Rh1-1D4 transgene is functional, because it rescues the phenotypic defects in ninaE I17 (20,21).…”
