Localization of a specific germ cell marker, DAZL1, in testicular germ cell neoplasias

Abstract: DAZ-like 1( DAZL1) is a germ cell-specific protein expressed in both male and female gonads. The DAZL1 gene, which maps to chromosome 3 in humans, is an autosomal homologue to the Deleted in AZoospermia ( DAZ) gene(s) located on the Y chromosome. We studied the expression of DAZL1 by means of immunohistochemistry in order to determine its distribution among testicular germ cell neoplasias and among the intratubular lesions in their vicinity. Our results demonstrated that expression of DAZL1 protein was consist… Show more

“…34 Indeed, the autosomal homologue of DAZ, DAZL1, is expressed in testicular germ cell tumours. 35 In conclusion, although our data do not show an effect of the Y chromosome background in testicular cancer in a cohort of English patients, it is necessary to study other populations where the incidence of TC is rising dramatically and where a Y chromosome haplogroup has been found to be associated with reduced testicular function, such as in Denmark. 28 …”

“…However, JKT-1 shows no signal for BMP-2 either, also excluding this possibility. We further demonstrate that JKT-1 lacks expression of the germ cell markers DAZL (Lifschitz-Mercer et al 2002) BOULE, and VASA (Ezeh et al 2005), raising the question as to whether JKT-1 can be regarded as a GCT line at all. Hence, the data presented here suggest that JKT-1, although initially clustering to embryonal carcinomas, does not express a convincing set of markers indicative for GCTs and germ cells.…”

“…Cells expressed the Sertoli cell marker Gata4 [17] and were negative for Dazl1 [24] and Vasa [25] (germ cells), Oct4 and Nanog (spermatogonial stem cells, [26]) and Hsd3β6 (Leydig cells, [27]) (Figure S1B). …”
