Localization in situ of the co‐stimulatory molecules B7.1, B7.2, CD40 and their ligands in normal human lymphoid tissue

Abstract: Functional interactions between B and T lymphocytes are known to depend on the expression of co-stimulatory molecules B7.1/CD80, B7.2/CD86 and their counter-receptors CD28 and CTLA4, as well as CD40 and its ligand CD40L. To study the role of these molecules in situ, an immunohistochemical analysis was carried out on normal human lymphoid tissue. In the germinal centers (GC), B7.1 and B7.2 were differentially expressed. In the dark zone, centroblasts were predominantly B7.1+, while centrocytes in the light zone… Show more

“…CD86 has an important role in the priming of naive T cells and activation of memory T cells (10,14,19,(27)(28)(29). Although activation of naive T cells requires strong stimulatory signals provided by APC (30), and activation of recently activated memory T cells can be elicited with anti-CD3 mAb alone (31), most memory T cells are still dependent on CD28 triggering for their activation (22).…”

“…In humans, resting monocytes constitutively express B7-2, whereas B7-1 expression is inducible in culture, particularly in the presence of IFN-g [21,22]. Tissue macrophages positive for both B7-1, B7-2 and CD40 have been identified in the interfollicular area of normal lymph nodes [23], but little is known concerning macrophages from other sites.…”

“…Slides were counterstained with hematoxylin and mounted in aqueous dry mounting medium. For detection of CD40L, an amplification step was performed with repeated incubation steps with biotinylated horse anti-mouse IgG and streptavidin/biotin-conjugated peroxidase (42).…”
