The germinal epithelium was eliminated in fifty post-pubertal albino rats by painting the ovarian surface with 10% tannic acid. Seven rats were treated bilaterally and forty-three unilaterally. The animals were killed at intervals ranging from a few hours to 469 days after the operation. Some germinal epithelium, usually abnormal in appearance, was present in eighteen out of the forty-three ovaries of the rats treated unilaterally, and in eight of fourteen ovaries of the animals dealt with bilaterally. The ovaries in the unilateral series were considerably reduced in volume. There was less involution in the bilaterally treated animals.Normal and atretic follicles in all stages of development were present up to 469 days after the elimination of the germinal epithelium.The total number of oocytes in the experimental animals was only slightly lower than normal, there being no significant difference (P > 0\m=.\4) between the numbers of oocytes in the experimental ovaries and in their controls. There were more normal large follicles in the control ovaries.The relevance of these findings to the question of post-pubertal oogenesis is discussed.

Primary oocytes exist in large numbers in the ovarian cortex of young animals. They themselves seem incapable of division and their abundance is owing to the earlier multiplication of the oogonia from which they have differentiated. As a feature of differentiation, the oocyte nucleus starts upon the early prophase changes of the first meiotic division, the chromosomes become somewhat condensed, and the nucleus then passes into the dictyate stage. Those oocytes that are not destined, as many are, for early degeneration remain in this stage until meiosis is suddenly resumed shortly before or soon after ovulation. The precise form assumed by the chromosomes in the dictyate stage is uncertain though they clearly lose their earlier partially condensed appearance. In oocyte nuclei in fish, amphibians, reptiles and birds, the chromosomes take on the form of fine long threads bearing numerous lateral loops, and are referred to as lampbrush chromosomes. Their special significance is still conjectural-they may play a part in yolk synthesis. Equivalent structures have yet to be demonstrated in mammalian oocytes. Recent observations of Ohno, Kaplan and Kinosita (i960) showed that the two X chromosomes in rat oocytes are isopycnotic, both at the first meiotic prophase and the second meiotic metaphase. This is in contrast to the positively heteropycnotic state of the XY bivalent in spermatocytes, as previously demonstrated by these workers (Ohno, Kaplan and Kinosita, 1957, 1958), and they suggest that the condition in spermatocytes represents an evolved mechanism that prevents crossing-over and ensures isolation of the femaledetermining chromosome from the male-determining chromosome. Developing rat ova have been studied with the electron microscope. Special attention was paid to relations of ova to the granulosa cells, the developmental stages of ovarian follicles, and the cytology of the unfertilized tubal ova. The relationship of the oocyte to the surrounding granulosa cells was found to change from one of a simple apposition of the plasma membranes to a complex interdigitation of microvilli from the ovular surface and processes from the granulosa cells extending into the matrix of the zona pelfucida. This complex interrelation is maintained until the formation of the first polar body is initiated. At this time no microvilli are found and the oolemma presents a gently undulating outline. Also at this time, a perivitelline space forms and the granulosa cell processes retract. In the unfertilized tubal ova no microvilli are present and the processes of the follicular cells are completely withdrawn.The cytoplasmic elements of the oocyte in various stages of development are described in some detail. Of particular interest is the change noted in position and degree of aggregation of the Golgi complex as maturation proceeds. Tile distribution and structural characteristics of the mitochondria, ergastoplasm, dense particles, and multivesicular bodies are described.
