Localisation of nucleolus-organising regions in interphase cells

Abstract: A technique based on the use of silver solutions, which selectively stains the nucleolus-organising regions (NORs) in chromosomes, was applied to interphase Ehrlich tumour cells. The results indicate that nucleolar fibrillar centres correspond to the NORs.

“…In addition, these observations have dem onstrated that the location of the silverstained spherules and the site of rDNA (as revealed by hybridization in situ) coincide with the site of the nucleolar fibrillar center. The data are in agreement with the role assigned to the fibrillar center, which has been identified as the site of the NOR in interphase nuclei (H ernandez-V erdun Goessens, et al, 1979). We have stained fibrillar centers as described by Hi rnandez-V erdun et al (1980) and Schwarzacher et al (1978) using a modification of the NOR-silver tech nique adapted for electron microscopy.…”

“…5). The components described above were also found in the fused nucle olus; i.e., a fibrillar center was formed as a result of the fusion of two fibrillar centers belonging to each small nucleolus and the associated electron-opaque fibrillar layer, which is known to be the site of rDNA transcription Stahl, 1978a, h, 1981;Goessens, 1979).…”

“…The secondary constrictions share common properties with the FCs. They are totally fibrous in ultrastructure (BRINKLEY 1965;RECHER et al 1972;GoESSENS and LEPOINT 1979) and have the capacity for selective staining with silver (HERNANDEZ-VERDUN et al 1978;SCHWARZACHER et al 1978;GoESSENS 1979). With gradual condensation of chromosomes accompanying concurrent NOR inactivation, the number of FCs decreases and at metaphase it corresponds to the number of silver-stained NORs (RoZAK et al 1986).…”

“…Some results have shown that fibrillar centers correspond to the nucleolusorganizer regions (NORs) containing the genes coding for ribosomal RNA (rRNA) [4,[10][11][12]. NORs can be selectively detected by silver staining methods [3,13].…”

“…In different types of nucleoli of somatic cells [3,4], cultivated cells [5,6] or sexual cells [7] a structure named the fibrillar center has been observed. It was first described by Recher et al [8) and reviewed by Goessens and Lepoint [9].…”
