Local antibody response in chickens: analysis of antibody synthesis to Newcastle disease virus by solid-phase radioimmunoassay and immunofluorescence with class-specific antibody for chicken immunoglobulins

Abstract: a stock B-i-type Lasota strain of ND containing 1084 50% effective lethal doses per ml; (ii) five were inoculated intratracheally (i.t.) and intranasally with 0.4 and 0.1 ml, respectively, of the same stock of virus diluted 1:10 in sterile phosphate-buffered saline (PBS); and (iii) five remained unexposed to NDV and served as controls. On days 4, 7, 10, 14, 21, and 28 after vaccination, saliva and blood were obtained from each bird. Thirty days after primary exposure to NDV, both groups were reinoculated i.t. w… Show more

“…Previous studies have shown the influx of plasma cells and T cells to the Harderian gland after infection with NDV 5 , 11 , 12 . In the Harderian gland, plasma cells have been shown to synthesize mainly IgA 5 , which has been detected as early as 4 dpi in the saliva of Leghorns 21 after infection with NDV. However, no IgA was detected in the lachrymal fluid of the challenged Fayoumis or Leghorns at 2 & 6 dpi (data not shown).…”

“…The host innate immune system rapidly produces cytokines, chemokines, and interferons in response to the virus, and triggers the adaptive immune response. Antibodies were detected as early as 4 days post infection in the saliva of Leghorns infected with lentogenic NDV [ 19 ]. Also, the lung is home to bronchus-associated lymphoid tissue (BALT), which plays a crucial role in antiviral immunity [ 20 ], and respiratory immune responses are induced in the lung [ 16 ].…”

“…Vaccination is the most accepted prevention and control strategy for combating ND in poultry worldwide. Commonly used vaccines are made with low virulence viruses, which provide both cellular and humoral immunity at a low cost, and inactivated oil-emulsion of the same viruses which confer a higher and long lasting humoral immunity [4][5][6]. In addition different recombinant NDV vaccines based on low virulence avian viruses (Herpervirus of Turkey, Fowlpox or low virulence NDV have been developed and used with great success experimentally.…”
