Local and sustained delivery of 5‐fluorouracil from biodegradable microspheres for the radiosensitization of glioblastoma

Abstract: This study demonstrates that biodegradable microspheres are efficient systems for drug delivery into the brain and may have future application in the treatment of brain tumors. Further studies are needed to confirm the potential of 5-FU-loaded microspheres for the radiosensitization of glioblastoma. [Please see editorial on pages 197-9, this issue].

“…The 5‐FU doses and injected volumes were the same regardless of tumor size and were determined from another pilot study of perioperative microsphere injection after tumor resection 27. No acute intracranial hypertension was observed despite the intratumoral injection of 2.5 mL of microsphere suspension, sometimes in large tumors.…”

“…In the blood samples, traces of 5‐FU were detected in some patients at Day 10 and Day 20. Compared with the 5‐FU kinetic study that was conducted after perioperative implantation of microspheres into the wall of resected tumor cavities,27 the concentrations of 5‐FU in the CSF and the systemic circulation were in the same range (from 0 ng/mL to 25 ng/mL). There was no correlation between the 5‐FU concentration and the neurologic side effects observed.…”

“…Second, heat and pressure are required to fabricate these depots in order to anneal and mold materials to a desired shape which may affect the stability of drugs especially gene, protein or hormone. Polymeric microspheres are also used as drug delivery systems for brain cancer chemotherapy [3,4]. However, fabrication of these particles is difficult to control size and porosity.…”

“…As an example of implantable devices, poly (lactic acid-co-glycolic acid) (PLGA) microspheres have been studied to deliver 5-fluorouracil (5-FU), which is a hydrophilic and antimetabolite drug used for cancer treatment [Menei 1999[Menei , 2004. After surgical resection of the tumor, 5-fluorouracil (5-FU) loaded PLGA microspheres are implanted intracranially to extend therapeutic duration and to avoid systemic toxicity around brain tissue.…”
