LncRNA AC245100.4 Binds HSP90 to Promote the Proliferation of Prostate Cancer

Abstract: Aim: To investigate the role and mechanisms of AC245100.4 in prostate cancer. Materials & methods: The expression and location of AC245100.4 were examined using real-time PCR and  in situ hybridization. Cell Counting Kit-8, clone formation, flow cytometry and in vivo assays were conducted to determine the role of AC245100.4. RNA antisense purification with mass spectrometry and RNA immunoprecipitation were performed to identify proteins that bind to AC245100.4. Western blotting was performed to quantify th… Show more

“…It is well known that LncRNAs mainly act as ceRNA to backout the inhibiting effects of miRNAs on their downstream targets 19,20 . SNHG1 was also reported to regulate miR-199a-3p and miR-337-3p/AKT2 axis, leading to promotion in PCa cells 11,12 . However, recently studies revealed that RNA binding protein plays a crucial role in LncRNA regulating network.…”

“…LncRNA prostate cancer antigen 3 (PCA3) has been approved by the U.S. FDA as a marker for the diagnosis of PCa, and has also been included in "Guidelines for Diagnosis and Treatment of Urological Diseases in China" 18 , announcing tremendous potential of lncRNA as a biomarker for the diagnosis and treatment of PCa, which is worthy of indepth study. Although several studies have proved that SNHG1 promoted PCa proliferation via serving as "miRNA sponeges" 11,12 , the functions and potential mechanisms in PCa remains unclear. We identified lncRNA SNHG1 was overexpressed in prostate cancer tissues and cells, and associated with patient's clinical characteristics.…”

“…For example, BiP surface translocation is required for different cotransporters, such as DNAJC3 ( Vig et al, 2019 ), Par-4 ( Cohen et al, 2013 ), and MTJ-1 ( Misra et al, 2005 ), in a context-dependent manner. Recent studies have indicated that lncRNAs can directly interact with the molecular chaperone HSP90, regulating signaling or protein stability by altering chaperone function ( Guo et al, 2018 ; Cui et al, 2020 ). Our data indicated that lnc-LTR5B binds to BiP in the cytoplasm.…”

“…On the contrary, a study conducted to determine the role of SNHG3 in breast cancer in a nude mouse model demonstrated that SNHG3 promotes cancer proliferation and metastasis by functioning as a miR-326 sponge [ 87 ]. The lncRNAs induce cancer invasion, proliferation, stemness, and metastasis by utilizing different mechanisms, i.e., by either promoting epithelial-to-mesenchymal transition (EMT) [ 88 ], regulating miRNAs expression profile via miRNAs sponging [ 89 ], modulating cell cycle including apoptotic pathways [ 90 ], interfering in mRNAs splicing [ 91 ], inducing resistance to different anti-cancer drugs [ 92 ], cellular metabolic reprogrammings [ 93 ], altering localization of proteins using direct binding [ 94 ], or regulating various associated metastatic signaling pathways [ 95 ].…”
