lncRNA Epigenetic Landscape Analysis Identifies EPIC1 as an Oncogenic lncRNA that Interacts with MYC and Promotes Cell-Cycle Progression in Cancer

Abstract: SUMMARYWe characterized the epigenetic landscape of genes encoding long noncoding RNAs (lncRNAs) across 6,475 tumors and 455 cancer cell lines. In stark contrast to the CpG island hypermethylation phenotype in cancer, we observed a recurrent hypomethylation of 1,006 lncRNA genes in cancer, including EPIC1 (epigeneticallyinduced lncRNA1). Overexpression of EPIC1 is associated with poor prognosis in luminal B breast cancer patients and enhances tumor growth in vitro and in vivo. Mechanistically, EPIC1 promotes c… Show more

“…Although great advances in early detection and treatment strategy lead to the steadily declined mortality, recurrence, and metastasis still occurred frequently and remained the major reasons for cancer deaths in patients with BC. Currently, a wide array of noncoding RNAs including microRNAs (miRNAs; Zare, Bastami, Solali, & Alivand, ), long noncoding RNAs (Wang et al, ) and circular RNAs (Zeng et al, ) have been identified to be involved in the tumorgenesis of BC as oncogenes or tumor suppressors.…”

“…Furthermore, many lncRNAs have important roles in various physiological and pathological processes . As to cancers, numerous lncRNAs have been shown to influence cell proliferation, cell cycle, cell apoptosis, cell senescence, cell migration and invasion, drug‐resistance, in vivo tumour growth and metastasis and so on . Therefore, several lncRNAs were identified to be potential therapeutic targets for various cancers .…”

“…LncRNA is a class of transcript with more than 200 nucleotides in length, but do not encode proteins . Aberrant expressions of lncRNAs have been revealed in various pathological status, particular in cancers . Furthermore, lncRNAs also play important roles in various pathophysiological processes, including cancers .…”
