lncRNA Chronos is an aging-induced inhibitor of muscle hypertrophy

Abstract: Neppl et al. identify a long noncoding RNA named Chronos whose expression increases with age and decreases in Akt-mediated growth. Inhibition of Chronos induces myofiber hypertrophy in vitro and in vivo, in part, through the epigenetic modulation of Bmp7 signaling.

“…The present data suggest that serum OIS1 may serve as an effective diagnostic biomarker in patients with HPV-positive cervical squamous cell carcinoma. Expression of lncRNAs is regulated by various factors, including aging (17), tobacco consumption (18) and alcohol abuse (19). In the present study, serum levels of OIS1 exhibited no significant association with patient age, smoking or alcohol consumption.…”

“…Recent findings indicate that denervation, immobilization, cancer cachexia, chronic kidney disease, fasting, and aging affect the expression of Pvt1, lncMUMA, Atrolnc-1, and MAR1 lncRNAs in the skeletal muscles [48][49][50][51]. In addition, the expression of Myoparr and Chronos is increased by surgical denervation and aging, respectively, and regulates skeletal muscle mass through BMP signaling [35,52], which antagonizes muscle atrophy [53,54]. Other lncRNAs, namely AK017368, Charme, and lnc-mg, are also involved in the regulation of skeletal muscle mass through multiple mechanisms [55][56][57].…”

“…Analogously to Mhrt, in skeletal muscle cells, the Gm17281 lncRNA, known as Chronos, inhibits hypertrophy through the Bmp7 signaling. Mechanistically, Chronos acts as an epigenetic repressor of Bmp7 cascade through the recruitment of EZH2 on the Bmp7 promoter (Neppl et al, 2017). In murine cardiac fibroblasts, a microarray analysis led to the identification of several lncRNAs dysregulated after pressure overload upon TAC.…”
