Liver-specific and proliferation-induced deoxyribonuclease I hypersensitive sites in the mouse insulin-like growth factor binding protein-1 gene

Abstract: The insulin-like growth factor binding protein-1 (IGFBP-1) gene is highly expressed in fetal, perinatal, and regenerating liver. Up-regulation is transcriptionally mediated in regenerating liver and occurs in the first few minutes to hours after partial hepatectomy. In transgenic mice a 970-bp region from ؊776 to ؉151 of the IGFBP-1 promoter was sufficient for tissue-specific and induced expression of the gene in fetal and hepatectomized livers. However weak and/or poorly regulated expression in some transgeni… Show more

“…Previous studies identified a number of hypersensitive sites around the IGFBP-1 gene that showed variable hypersensitivity correlating with the proliferative state of the liver (9). We found that the same hypersensitive sites at Ϫ5000, Ϫ3100, and Ϫ100 to Ϫ300 that showed increased hypersensitivity posthepatectomy in normal mice showed a relative decrease in hypersensitivity in hepatectomized IL-6 Ϫ/Ϫ livers (data not shown).…”

“…3B. Oligonucleotide sequences for E2, c-Fos STAT3, C/EBP␣/␤, TTR-HNF-3 (Ϫ111/Ϫ85), and Ϫ3118/Ϫ3094 were described by Crissey et al (9). The sequences for AP-1 and HNF4 are 5Ј-GATCCTTGAGTCACATCGATTGAG TCACG-3Ј and 5Ј-GGAAAGGTCCAAAGGGCGCCTTG-3Ј, respectively.…”

“…In the present study, we characterized the elements in the promoter of human P4H␣(I). We found that the E-box-like sequence (CACGGG), which has been shown to bind upstream stimulatory factors (USF) (25)(26)(27), in the position of bp Ϫ135 is vital for P4H␣(I) transcription. Upstream stimulatory factor (USF1/USF2) appears to be the transcription factor that controls the basal P4H␣(I) expression.…”
