Liver-derived human mesenchymal stem cells: a novel therapeutic source for liver diseases

Abstract: Mesenchymal stem cells (MSCs) represent an attractive cell type for research and therapy due to their ability to proliferate, differentiate, modulate immune reactions, and secrete trophic factors. MSCs exist in a multitude of tissues, including bone marrow, umbilical cord, and adipose tissues. Moreover, MSCs have recently been isolated from the liver. Compared with other MSC types, liver-derived human MSCs (LHMSCs) possess general morphologies, immune functions, and differentiation capacities. Interestingly, L… Show more

“…Although the current study is not the first attempt at characterizing the L-MSCs, (48) it differs from and extends the results of the previous ones significantly. Mainly, we investigated MSCs obtained from primary adult human liver tissue, rather than infant liver, (49) cryopreserved hepatocytes, (50) or liver allograft perfusate as a source.…”

“…We now know that the assumption that MSCs are derived from the connective tissue (stroma) of marrow or the stroma of other tissues is not correct. This assumption became more untenable when reports came forth of MSCs isolation from almost every tissue of the body: Fat, skin, heart, muscle, liver, etc . The landmark and comprehensive report of Crisan et al clearly documents that MSCs are derived from perivascular cells (pericytes) from a number of human tissues.…”

“…Thus, it is plausible that the increased liver colonization is regulated by the MSX1-ZEB1 axis. Furthermore, the hepatic microenvironment provides a favorable milieu for mesenchymal stem cells, which likely play a role in the promotion of liver regeneration (Liu et al, 2015; Wang et al, 2016). …”
