Live single cell mass spectrometry reveals cancer‐specific metabolic profiles of circulating tumor cells

Abstract: Recently, there has been increased attention on the analysis of circulating tumor cells (CTCs), also known as liquid biopsy, owing to its potential benefits in cancer diagnosis and treatment. Circulating tumor cells are released from primary tumor lesions into the blood stream and eventually metastasize to distant body organs. However, a major hurdle with CTC analysis is their natural scarcity. Existing methods lack sensitivity, specificity, or reproducibility required in CTC characterization and detection. He… Show more

“…Yet, it is the development of single-cell technologies during the past decade that has enabled a comprehensive interrogation of CTCs from patients with various cancer types. Combined with next-generation sequencing (NGS) and mass cytometry technologies, it is now possible to characterize the genome, transcriptome, methylome [18], and proteome [19] of individual cells. The recent development of a single-cell RNA-seq platform based on a hydrodynamic barcoding technique (Hydro-Seq) also shows a promising efficiency for wholetranscriptome analysis of CTCs [20].…”

“…Another similar integration with 3D holography has allowed for achieving absolute quantitation of cellular metabolites on the subcellular level 16 . Additionally, integration with flow cytometry has allowed for the uncovering of metabolic biomarkers in single circulating tumor cells of neuroblastoma cancer patients 17,18 .…”

“…By exploiting this property, CTCs from several solid cancer models have been successfully distinguished from surrounding blood cells 83 . Similarly, a mass spectrometry approach combined with microfluidics platform has recently been used to efficiently discriminate colorectal cancer patients from those with gastric cancer, based on their CTCs metabolomic profile 84 …”
