Liquiritin Alleviates Pain Through Inhibiting CXCL1/CXCR2 Signaling Pathway in Bone Cancer Pain Rat

Abstract: Bone cancer pain (BCP) is an intractable clinical problem, and lacked effective drugs for treating it. Recent research showed that several chemokines in the spinal cord are involved in the pathogenesis of BCP. In this study, the antinociceptive effects of liquiritin, which is an active component extracted from Glycyrrhizae Radix, were tested and the underlying mechanisms targeting spinal dorsal horn (SDH) were investigated. The BCP group displayed a significant decrease in the mechanical withdrawal threshold o… Show more

“…The establishment of a BCP rat model was done as described in previous studies [ 36 , 37 ]. Briefly, the rats were anesthetized via intraperitoneal injection with sodium pentobarbital (50 mg/kg).…”

“…Although no statistical method was used to determine the sample number in advance, we determined the required sample size for each group with reference to the previous literature in this field. The behavioural experiments were 6 rats per group (Ni et al, 2019), 4 rats per group for western blot (He et al, 2020) and immunofluorescence (Ni et al, 2020), 5 rats per group for real-time quantitative PCR (Mohamed et al, 2020) and enzyme-linked immunosorbent assay (Zhang et al, 2019) and 10 rats per group for computed tomography (CT) reconstruction and histological analysis of bone (Xu et al, 2019). Referring to the previous literature (Ma et al, 2020), the animals were simply randomly grouped.…”

“…Our study demonstrated that upregulation of miR-532-5p suppressed CI/RI through the CXCL1/CXCR2 pathway. CXCL1 is known to be a crucial mediator in inflammation and organ injury [ 27 ], and CXCR2 is the downstream protein of CXCL1 [ 28 , 29 ]. In a previous study, CXCL1/CXCR2 pathway could lead to the injury of brain tissues [ 15 ].…”
