Liquid‐to‐solid ratio control as an advanced process control solution for continuous twin‐screw wet granulation

Abstract: Assuring compliance of intermediate and final quality attributes in a continuous pharmaceutical manufacturing campaign is of utmost importance. Application of corrective actions might be required in real‐time. This work exemplifies the steps needed to identify a linear pulse transfer function for the dynamic behavior of the granule liquid‐to‐solid ratio (%w/w) at the end of the granulation unit of a commercial ConsiGmaTM‐25 production line. Near‐infrared spectroscopy was used to monitor the granule composition… Show more

“…By in-line NIR spectroscopy monitoring, the L/D ratio of the wet granule based on a PLS regression model and an automatic supervisory controller was established for TSWG. When inducing artificial disturbances, it correctly follows the dynamic set point and obtains an RMSE of 0.25% w/w relative to the setpoint [76]. Torque is an important PV in the TSWG process, and the feasibility of the APC approach by adjusting the torque was investigated.…”

“…Hence, it has been successfully applied in industry, as summarized in [39]. Hitherto, it was applied in a manifold of technical and non-technical areas, while being recently introduced in the pharmaceutical indus-try [40]. Stability and feasibility of this algorithm have been given in [41], [42] and robustness has been discussed in [43].…”

“…Unlike other academic MPC strategies, the EPSAC approach is based on input-output filtering techniques, making it thus very attractive for industrial applications. The simplicity of the methodology allowed its implementation on FPGAs platforms [9] or dSpace platforms [10] and other real time operating environments [11], [12].…”
