Liquid chromatography/time-of-flight mass spectrometry with high-speed integrated transient recording

Abstract: High-performance liquid chromatography (HPLC) has been interfaced to a time-of-flight mass spectrometer. The interface is a continuous flow probe and ions are desorbed from the liquid matrix by energetic ion bombardment. Pulsed and delayed ion extraction from the source permits the use of broad ionization times, results in the production of analog signals in each time-of-flight cycle, and provides both energy and spatial focusing. A high-speed integrated transient recording system has been developed and is als… Show more

“…In the 2000s, technical advances led to the development of high-resolution mass spectrometry systems. The first commercial Fourier transform ion cyclotron resonance (Marshall et al, 1998), orbitrap (Hu et al, 2005), and time-of-flight (Emary et al, 1990) mass spectrometers, coupled with HPLC, were introduced into the marketplace. Unit mass resolution, as is obtained with quadrapoles, was replaced by highresolution (4140 000) mass analyses, a very advantageous feature for the analysis of complex biological matrices.…”

“…In their case, the injection came from an atmospheric-pressure plasma ion source (8). Unfortunately, the work went largely unnoticed by most, so the technique was independently reinvented by many groups 20-25 years later using various types of continuous ion sources (9)(10)(11)(12)(13)(14)(15)(16). The earliest of these is the most relevant to the present discussion: Dodonov et al were the first to couple an ESI source to an orthogonal injection TOF instrument which made it particularly useful for observing biomolecules.…”

“…The earliest of these is the most relevant to the present discussion: Dodonov et al were the first to couple an ESI source to an orthogonal injection TOF instrument which made it particularly useful for observing biomolecules. They were also the first to use a reflecting geometry with orthogonal injection (9 [17][18][19] which improved resolution Their ion source stemmed from Russian (10), electron ionization (11,14), liquid SIMS (12,15), a cluster ion source (13), atmospheric-pressure ionization (16,17), and inductively coupled plasma (20).…”

“…Data recording also appears to be a problem in other timeof-flight mass spectrometer systems. 10,11 Various solutions for the spectral recording system have been attempted. The initial systems utilized photographic techniques.…”

“…Several sophisticated electronic systems have been developed to take advantages of this facility. 2,7,10 Initially, hard copy of the mass spectra could be obtained from these systems. The rapid progress in computer technology in recent years has stimulated attempt to develop computerization of the spectrum recording systems.…”
