Lipoxygenase Inhibitors Nordihydroguaiaretic Acid and Fungus Lecanicillum lecanii Extract Induce Death of Lymphoid Leukemia Cells

Abstract: We studied the effect of lipoxygenase inhibitors nordihydroguaiaretic acid (NDGA) and fungus Lecanicillum lecanii extract on lymphatic leukemia P388 cells. The cells grown in the abdominal cavity of DBA2 mice for 7 days were transferred into a nutrient medium. The effect of lipoxygenase inhibitors was evaluated by changes in cell number, trypan blue staining, nucleus damage, and changes in cell distribution by DNA content after 22-h incubation. NDGA and fungus extract induced apoptotic death of lymphatic leuke… Show more

“…It is widely distributed in the warm deserts of North American and Argentina [2], and known to attain ages of several thousand years [3]. Apart from the pharmacological use of L. tridentata in traditional system of medicine for diverse ailments [4], it contains the potent antioxidant phenolic lignan / phenylpropanoids, nordihydroguaiaretic acid (NDGA) [4][5][6][7][8][9] which exerts in vitro anti-cancer effects [10,11].…”

“…The nordihydroguaiaretic acid (NDGA) is the most abundant and studied metabolite of LT [ 22 ]. NDGA is a potent inhibitor of lipoxygenase and cyclooxygenase pathway [ 26 ]. Tea brewed of LT was used to treat digestive disorders, rheumatism and most common colds [ 23 ].…”

“…Tea brewed of LT was used to treat digestive disorders, rheumatism and most common colds [ 23 ]. In 2018, Chan et al reported that NDGA protect against obesity in vivo model [ 26 ]. LT is a rich source of metabolites including phenolic, terpenoids, saponins, and odorous vinyl ketones ( Figure 1 ) [ 27 ].…”

“…Disruption of the actin cytoskeleton in association with the activation of stress-activated protein kinases has also been observed (Seufferlein et al, 2002). NDGA, along with the extract from the fungus Lecanicillum lecanii, inhibits the growth of lymphatic leukemia cells by inducing nuclear damage leading to reduced DNA content in the cells (Bibikova et al, 2017). Besides, the compound causes cell death in AML cell lines as well as in patient samples via inhibition of AKT phosphorylation, suggesting that the extrinsic and the mitochondrial apoptotic pathways are not essential for cell death (Mak et al, 2007).…”
