Nanoparticles with pH-sensitive behavior may enhance the success of chemotherapy in many cancers by efficient intracellular drug delivery. Here, we investigated the effect of a bioactive surfactant with pH-sensitive properties on the antitumor activity and intracellular behavior of methotrexate-loaded chitosan nanoparticles (MTX-CS-NPs). NPs were prepared using a modified ionotropic complexation process, in which was included the surfactant derived from N α ,N ε -dioctanoyl lysine with an inorganic lithium counterion. The pH-sensitive behavior of NPs allowed accelerated release of MTX in an acidic medium, as well as membrane-lytic pH-dependent activity, which facilitated the cytosolic delivery of endocytosed materials. Moreover, our results clearly proved that MTX-CS-NPs were more active against the tumor HeLa and MCF-7 cell lines than the free drug. The feasibilty of using NPs to target acidic tumor extracellular pH was also shown, as cytotoxicity against cancer cells was greater in a mildly acidic environment. Finally, the combined physicochemical and pH-sensitive properties of NPs generally allowed the entrapped drug to induce greater cell cycle arrest and apoptotic effects. Therefore, our overall results suggest that pH-sensitive MTX-CS-NPs could be potentially useful as a carrier system for tumor and intracellular drug delivery in cancer therapy.

Антимитотический агент комбретастатин А4 (СА-4) сравнительно недавно предложен в качестве антиваскулярного средства для противоопухолевой терапии. С целью уменьшения системной токсичности за счет применения в виде липосомальных препаратов, в данной работе синтезированы новые липофильные пролекарства -олеоильные производные СА-4 и его 4-арилкумаринового аналога (ArC): СА4-Ole и ArC-Ole, соответственно. Показано, что липосомы среднего диаметра 100 нм, полученные на основе яичного фосфатидилхолина и фосфатидилинозита из пекарских дрожжей, количественно включают до 15 моль-% СА4-Ole или 7 моль-% ArC-Ole. Также получены липосомы с пролекарствами, оснащённые тетрасахаридным лигандом селектинов, а именно сиалил-Lewis X (SiaLe X ), с целью адресной доставки к эндотелию формирующейся de novo сосудистой системы. Противоопухолевые свойства in vivo изучали на модели медленно растущего рака молочных желез (РМЖ) мышей. В используемых дозировке (22 мг/кг) и протоколе введения (4 раза с интервалом в одну неделю, начиная с появления пальпируемых опухолей) цитостатик СА-4 не оказал противоопухолевого действия, более того, даже стимулировал рост РМЖ. Липосомальные формы CA4-Ole не оказывали стимулирующего действия на рост опухолей. Однако, для получения выраженного противоопухолевого эффекта, очевидно, необходимо увеличить кратность введения препаратов липосом. Новый антимитотический агент ArC показал меньший на порядок уровень цитотоксичности in vitro в культуре клеток карциномы молочной железы человека. Тем не менее, при испытаниях на мышиной модели РМЖ полученный аналог показал противоопухолевый эффект в двухкратной эквивалентной дозе по СА-4. Результаты показывают перспективность применения ArC-Ole в липосомах, оснащённых SiaLe X : препарат частично подавлял рост опухоли уже после второго введения. A number of antiangiogenic drugs have been approved by the Food and Drug Administration which are used in cancer therapy, and variety of other agents in several stages of clinical development or in preclinical assessment. Among these, combretastatin A4 (CA‐4) is an under‐researched inhibitor of angiogenesis that shows potential activity in the treatment of advanced tumors with migration capacity. However, its clinical application has been limited due to poor water solubility, low bioavailability, rapid metabolism, and systemic elimination. During the last decade, numerous investigations have been done to overcome these problems by using different CA‐4 delivery systems or developing produgs of CA‐4 or its structural analogs. Nevertheless, these strategies could not be efficient out of the undesired side effects on normal tissues. Nanoliposomal CA‐4 not only benefits from the advantage of using liposomal drugs as opposed to free drugs but also can accumulate in the tumor site via specific targeting ligands, which leads to efficient targeting and enhancement of bioavailability. To the best of our knowledge, we consider an important attempt to understand different factors that might influence the CA‐4 loading and release pattern of liposomes and the consequent results in tumor therapy. In this review, we shed light on various studied liposomal CA‐4 formulations showing application thereof in cancer treatment.
