Liposomes for Specific Depletion of Macrophages from Organs and Tissues

Abstract: A liposome mediated macrophage "suicide" approach has been developed, based on the liposome mediated internalization of the small hydrophilic molecule clodronate in macrophages J Leukoc Biol 62:702, 1997. This molecule has a very short half life when released in the circulation, but does not easily cross phospholipid bilayers of liposomes or cell membranes. As a consequence, once ingested by a macrophage in a liposome encapsulated form, it will be accumulated within the cell as soon as the liposomes are digest… Show more

“…Phagocytic uptake of liposome-encapsulated clodronate (Clo-Lipo) is a well-established method for specific in vivo ablation of macrophages (24). This reagent was effective in specifically depleting circulating axolotl monocytes and tissue resident macrophages on i.v.…”

“…to selectively ablate macrophages in a variety of systems, including the inner ear (Van Rooijen and Sanders 1994;van Rooijen et al 1996;van Rooijen and Hendrikx 2010;Warchol et al 2012). We first examined the effect of LC on macrophage survival by treating utricles from CX3CR1 GFP/+ mice, which express GFP exclusively in macrophages in the inner ear (Hirose et al 2005).…”

“…The first involved the intravenous injection of liposomeencapsulated clodronate, an intracellular toxin taken up by monocyte/macrophages that has been used extensively to study the effects of monocyte depletion in mice. 34,44 To study the contribution of monocytes in HIT, we depleted monocytes with clodronate liposomes in transgenic Fc␼RIIA ĎŠ /hPF4 ĎŠ mice. We used flow cytometry to measure the relative number of cells expressing the monocyte differentiation marker CD115 as a proportion of total WBC characterized by expression of CD45.…”
