Liposome solubilization induced by complexation with dimeric β-cyclodextrin

Abstract: Dimeric b-cyclodextrins (b-CD) were prepared from the reaction of native b-CD with epichlorohydrin under basic conditions, and the effects on the diacetylene (DA) and polydiacetylene (PDA) liposomes have been investigated. Vesicular DA was solubilized in the presence of dimeric b-CD with the consequent inhibition of polymerization. The result is attributed to the formation of a complex between dimeric b-CD and DA liposomes, and it is clearly differentiated from that of monomeric b-CD. Furthermore, the ordered … Show more

“…Following this, 6 PCD β-CD (or 3 PCD β-CD) and HEPES buffer solution (5 mM, pH 8.0) was added to yield a total concentration of 1 mM. The samples were then heated at 80 °C for 15 min and probe sonicated for 12.5 min using a Sonics VC-505 instrument at 40% of the power used 41 . The resulting solution was filtered through a 0.8 μm filter (Satorius, Minisart), and the milky filtrate was cooled overnight at 4 °C.…”

“…PCDA (4.48 mg, 0.8 mmoL) was dissolved in 1 mL of chloroform, and the organic solvent was removed by purging with N 2 to generate a thin lipid film on the glass surface . Then, SOM‐PCDA and HEPES buffer solution (5 × 10 −3 m , pH 8.0) were added for a total lipid concentration of 1 × 10 −3 m .…”
