Lipocalin 2 Diminishes Invasiveness and Metastasis of Ras-transformed Cells

Abstract: Lipocalin 2, an iron-siderophore-binding protein, converts embryonic kidney mesenchyme to epithelia. We found that lipocalin 2 could also convert 4T1-Ras-transformed mesenchymal tumor cells to an epithelial phenotype, increase E-cadherin expression, and suppress cell invasiveness in vitro and tumor growth and lung metastases in vivo. The Ras-MAPK pathway mediated the epithelial to mesenchymal transition in part by increasing E-cadherin phosphorylation and degradation. Lipocalin 2 antagonized these effects at a… Show more

“…Hanai et al (2005) recently showed that NGAL has the ability to reverse this epithelial-mesenchymal transition, thereby diminishing invasiveness of cancer cells. Our results show that in pancreatic cancer, NGAL is predominantly expressed in the early dysplastic lesions as well as in the well-differentiated carcinomas (preserve ductal architecture); with almost no expression in the poorly differentiated tumours (have a predominantly mesenchymal appearance).…”

“…The involvement of lcn2 in the phenotypic change of cell morphology has been previously reported. lcn2 was an endogenous epithelial inducer (Yang et al, 2002) and stimulated the epithelial phenotype of transformed cells (Hanai et al, 2005;. lcn2 also promoted tubulogenesis by regulating epithelial morphogenesis (Gwira et al, 2005).…”

“…Locally, even higher levels may be achieved in pathologic conditions. At 50 g/ml, LCN2 induced kidney epithelia differentiation (Yang et al, 2002), epithelial transition of cancer cells (Hanai et al, 2005), and apoptosis of pro-B-cells (Devireddy et al, 2005) in an iron-dependent manner (Mori and Nakao, 2007). Purified GST protein (Fig.…”

“…LCN2 was found to be highly expressed after malignant transformation of the breast, lung, colon, and pancreatic epithelia by immunohistochemistry (39). More recently, it has been described that LCN2 decreases the invasiveness and metastasis of Ras-transformed cells (40). BMP4 is a member of the bone morphogenetic protein family, which is part of the transforming growth factor-␤ superfamily.…”
