Lipid raft disruption prevents apoptosis induced by 2-chloro-2′-deoxyadenosine (Cladribine) in leukemia cell lines

Abstract: These results indicated that lipid rafts partly contributed to 2CdA-induced apoptosis by regulating Ca 2+ influx via the plasma membrane.3

“…Our data demonstrated that cholesterol depletion from membranes inhibits fluid flow induced Ca 2þ i mobilization, suggesting cholesterol in cell membranes may be involved in fluid flow induced calcium signaling. These results are consistent with previous studies, in which cholesterol depletion inhibits hyaluronan-mediated and caldribine-induced Ca 2þ i mobilization in endothelial cells (Singleton and Bourguignon, 2004) and leukemia cells (Takahashi et al, 2006), respectively. However, it is the first report that mechanical stimulation (oscillatory fluid flow) induced Ca 2þ i was inhibited by MbCD.…”

“…In acute promyelocytic leukemia and lymphoblastic leukemia cells, changes in intracellular calcium levels occurred after treatment with all the tested derivatives 31 . Studies suggest that lymphocytes express L-type Ca 2+ channels that are activated in a non-voltage-gated manner 32 . The obtained data revealed that compounds 1 and 2 are potent to activate caspase 3, the enzyme which is involved in proteolysis of numerous proteins (e.g.…”

“…Akt/PKB [62]. The effects of raft disruption on pro-apoptotic signaling have also been demonstrated in B cells, where lipid rafts play a crucial role in CD20-induced caspase activation [63] or in leukemic cells where lipid rafts disruption prevents cladribine induced apoptosis [64]. Finally, by using MβCD and filipin, it was also demonstrated a role for lipid rafts in cell sensitization to cathepsin B-dependent apoptosis via lipopolysaccaride [65].…”
