Lipid mediator n‐3 docosapentaenoic acid‐derived protectin D1 enhances synaptic inhibition of hippocampal principal neurons by interaction with a G‐protein‐coupled receptor

Abstract: Epilepsy is a severe neurological disease manifested by spontaneous recurrent seizures due to abnormal hyper‐synchronization of neuronal activity. Epilepsy affects about 1% of the population and up to 40% of patients experience seizures that are resistant to currently available drugs, thus highlighting an urgent need for novel treatments. In this regard, anti‐inflammatory drugs emerged as potential therapeutic candidates. In particular, specific molecules apt to resolve the neuroinflammatory response occurring… Show more

“…When mice were treated with PD1 n-3 DPA using a therapeutic paradigm, disease severity, including the expression of pro-inflammatory cytokines and the frequency and duration of epileptic seizures, was significantly reduced [ 171 ]. Furthermore, recent studies demonstrate that PD1 n-3 DPA is also able to increase the inhibitory drive onto the perisomatic region of the pyramidal neurons thereby limiting neuronal excitability [ 172 ].…”

“…PD1 n‐3DPA exerts its anti-inflammatory effects via increasing the inhibitory transmission, at the presynaptic site of inhibitory synapses. PD1 n‐3DPA also exhibits anti‐seizure and neuroprotective effects by modulating the synaptic inputs [243] .…”
