Lipid and fatty acid analysis of fresh and frozen-thawed immature and in vitro matured bovine oocytes

Abstract: ResearchThe lipid content and fatty acid composition of fresh immature and in vitro matured bovine oocytes cultured in media with or without serum, and also those of frozen-thawed immature oocytes were analysed. All oocytes were ranked (A or B) on the basis of their cytoplasmic quality. Fatty acid composition (mol %; w/w) in the total lipid fraction was analysed by gas chromatography. Triglyceride, total cholesterol, phospholipid (phosphocholine-containing phospholipid) and non-esterified fatty acid contents o… Show more

“…É possível também a visualização de folículos primordiais, primários, secundários e do epitélio germinativo. (Didion et al, 1990), pois a redução da temperatura ocasiona a alteração da morfologia, estrutura e distribuição celular destas gotículas, consequetemente ocasionando a rupture do citoesqueleto (Kim et al, 2001). Também existe a circunstância de que a eficácia do resfriamento do oócito depende da sua fase de desenvolvimento (Ferreira et al, 2001).…”

“…Most information regarding lipid structural analysis from oocytes and embryos has been obtained by staining methods, such as Nile Red, which quantify cytoplasmic lipids in general and gas chromatography (GC) that quantify by fatty acid (FA) residues extracted from the total sample lipid content (BARCELÓ-FIMBRES;SEIDEL JR., 2011;KIM et al, 2001;LAPA et al, 2011;MATORRAS et al, 1998).…”

“…However, techniques that involve the manipulation of oocytes and/or embryos may themselves be detrimental to embryo viability and subsequent development (ALLEN, 2005;COUTINHO DA SILVA, 2008;SQUIRES et al, 2003). Quality of the equine embryo has a major effect on pregnancy rates, and embryo cryopreservation (ALLEN, 2005;LAGARES et al, 2009;SQUIRES, 2005).…”

“…Dietary fatty acids altered the composition of fatty acids in the cumulus cells, granulosa cells and oocytes that may also have consequences on oocyte quality (Kim et al, 2001). For example, in ewes, the number and quality of oocytes was increased when animals were fed a diet enriched in PUFAs (saponified fish oil) 74.3% v. 57%; (Zeron et al, 2002).…”
