Linking RNA Processing and Function

Abstract: RNA processing is critical for eukaryotic mRNA maturation and function. It appears there is no exception for other types of RNAs. Long noncoding RNAs (lncRNAs) represent a subclass of noncoding RNAs, have sizes of >200 nucleotides (nt), and participate in various aspects of gene regulation. Although many lncRNAs are capped, polyadenylated, and spliced just like mRNAs, others are derived from primary transcripts of RNA polymerase II and stabilized by forming circular structures or by ending with small nucleolar… Show more

“…In addition, the cohorts differed in RNA-sequencing protocols, with total RNA sequencing for FF_RP and FFPE_Bx versus poly(A)-enriched RNA libraries for TCGA_PRAD and DKFZ_RP. Total RNA sequencing enables analysis of all ncRNA families, whereas poly(A)-sequencing captures only RNA with a poly(A) tail, such as mRNA and some long ncRNA families [ 24 ]. The DKFZ_RP cohort also allowed us to characterize fusions from the perspective of early-onset PCa and to assess the prognostic potential of gene fusions in an early-onset PCa cohort.…”

“…Another significant difference is that mRNAs in prokaryotes tend to be polycistronic mRNAs; by contrast, eukaryotic mRNAs are usually monocistronic mRNAs, Promoter: DNA region that defines where the transcription of a gene by although this is not always the case [21,22]. Importantly, transcription and translation often occur simultaneously in prokaryotes, but in eukaryotes the RNA is transcribed in the nucleus and translated in the cytoplasm [23].…”

“…Compelling evidence has illustrated that during the pathogenesis of IDD, long noncoding RNAs (lncRNAs) are pivotal molecules by functioning in nucleus pulposus cells (NPCs) [ 8 , 9 ]. LncRNAs are long RNA transcripts with size over 200 nucleotides (nt) and incapable of encoding proteins [ 10 , 11 ]. However, the amounts of researches studying the role of lncRNAs in IDD remain rare.…”
