Linkage of E2F1 transcriptional network and cell proliferation with respiratory chain activity in breast cancer cells

Abstract: Mitochondria are multifunctional organelles; they have been implicated in various aspects of tumorigenesis. In this study, we investigated a novel role of the basal electron transport chain (ETC) activity in cell proliferation by inhibiting mitochondrial replication and transcription (mtR/T) using pharmacological and genetic interventions, which depleted mitochondrial DNA/RNA, thereby inducing ETC deficiency. Interestingly, mtR/T inhibition did not decrease ATP levels despite deficiency in ETC activity in diff… Show more

“…E2F1 also functions as a connector and coordinator between cell proliferation and metabolic pathways in mitochondria. Mitochondria are multifunctional organelles, which are suggested to be actively involved in various aspects of tumorigenesis (17). It has been reported that E2F1 enhances glycolysis through suppressing the transcription of Sirt6 in PC3 prostate cancer and UMUC3 bladder cancer cells (18).…”

“…All data are representative of three independent experiments with measurements in triplicate for each experiment. Anchorage-independent cell growth was assayed as described previously [43] with slight modifications. In brief, a single-cell suspension was prepared from monolayer culture by treatment with TrypLE TM Express (Thermo Fisher Scientific) and cultured at a density of 1.0 9 10 4 cells per well in polyHEMA-coated six-well plates with growth medium supplemented with 1.0% methylcellulose.…”

“…Some evidences show that mitochondrial function, in turn, also impacts E2F1 activity. For instance, inhibition of ATP synthase or of the electron transport chain leads to the downregulation of E2F1 activity and to cell cycle arrest ( 79 , 80 ). On the other hand, mitochondrial ROS production can promote E2F1-mediated apoptosis ( 81 , 82 ).…”
