Arthrobacter simplex is extensively used for cortisone acetate (CA) biotransformation in industry, but the Δ 1dehydrogenation molecular fundamental remains unclear. Herein, the comparative proteome revealed several proteins with the potential role in this reaction, which were mainly involved in lipid or amino acid transport and metabolism, energy production and conversion, steroid degradation, and transporter. The influences of six proteins were further confirmed, where pps, MceG A , yrbE4A A , yrbE4B A , and hyp2 showed positive impacts, while hyp1 exhibited a negative effect. Additionally, KsdD5 behaved as the best catalytic enzyme. By the combined manipulation in multiple genes under the control of a stronger promoter, an optimal strain with better catalytic enzyme activity, substrate transportation, and cell stress tolerance was created. After biotechnology optimization, the production peak and productivity were, respectively, boosted by 4.1-and 4.0-fold relative to the initial level. Our work broadens the understanding of the Δ 1 -dehydrogenation mechanism, also providing effective strategies for excellent steroid-transforming strains.

Both the bacterial flagellum and the evolutionary related injectisome encoded on the Salmonella pathogenicity island 1 (SPI-1) play crucial roles during the infection cycle of Salmonella species. The interplay of both is highlighted by the complex cross-regulation that includes transcriptional control of the flagellar master regulatory operon flhDC by HilD, the master regulator of SPI-1 gene expression. Contrary to the HilD-dependent activation of flagellar gene expression, we report here that activation of HilD resulted in a dramatic loss of motility, which was dependent on the presence of SPI-1. Single cell analyses revealed that HilD-activation results in a SPI-1-dependent induction of the stringent response and a pronounced decrease of proton motive force (PMF), while flagellation was not affected. We further found that activation of HilD enhanced the adhesion of Salmonella to epithelial cells. A transcriptome analysis revealed a concomitant upregulation of several adhesin systems, which when overproduced, phenocopied the HilD-induced motility defect. We propose that a combination of SPI-1-dependent depletion of the PMF and upregulation of adhesins upon HilD-activation allows flagellated Salmonella to rapidly modulate their motility during infection, thereby enabling efficient adhesion to host cells and delivery of effector proteins.

Many bacteria move in liquid environments using a macromolecular propeller, the flagellum. A type-III secretion system (T3SS) at the base of the flagellum and the related injectisome of Gram-negative pathogens are required to build and operate these sophisticated nanomachines. The flagellar structure can be divided into three main parts:(a) a cell envelope-anchored basal body (consisting of the C-ring, the flagellar-specific T3SS, the MS-ring, the rod and the LP-ring), (b) a hook and, (c) a filament. Secretion of a variety of different T3SS substrate proteins occurs from the cytoplasm to the distal end of the growing structure in a highly regulated and sequential manner (Chevance & Hughes, 2008;Diepold & Wagner, 2014;Wagner et al., 2018).
