Linkage between a muscle-specific CK gene marker and &OV0312;O2max in the HERITAGE Family Study

Abstract: The present results provide further support for the notion that the CKMM gene, or some gene in close linkage disequilibrium with it, may contribute to individual differences in the VO2max response to endurance training.

“…Recently interdependence between the CK-MM A/G polymorphism and individual differences in the expression of the physical qualities of a person has been shown in several studies (17,18). Association of the CK-MM A/G polymorphism with the increment in aerobic power after aerobic physical training of subjects not engaged in sports has been demonstrated (13,16,19).…”

“…It is believed that the A/G polymorphism is also associated with different CK-MM activities in myocytes (13). The frequency of the minor G allele in this type of polymorphism varies in the range from 15% in the Chinese population to 29% -35% in Caucasians and 32% in white Americans (14)(15)(16). Recently interdependence between the CK-MM A/G polymorphism and individual differences in the expression of the physical qualities of a person has been shown in several studies (17,18).…”

“…Although numerous studies have reported a relationship between these two items [9][10][11][12][13][14], it remains controversial. Furthermore, mitochondrial DNA (mtDNA) [15,16], the creatine kinase gene [17][18][19] and the Na ϩ -K ϩ -ATPase ∂2 gene [20] have been studied as genes that may dictate individual differences in endurance capacity or trainability. mtDNA is a double-stranded, circular molecule of 16,569 bp within the mitochondria [21] and has many more polymorphisms than seen in nuclear DNA [22].…”

“…A polymorphism in the CK-M gene was detected by chain polymerase reaction and DNA digestion with the endonuclease enzyme of NcoI restriction. The allele with site susceptible to digestion for NcoI was designated as 985+185 pb allele, while the allele without the site for NcoI restriction was designated as 1170 pb allele (69) . This variable defined as NcoI is located in the region 3' of the gene (70) .…”
