bFoods and related processing environments are commonly contaminated with the pathogenic Listeria monocytogenes. To investigate potential environmental reservoirs of Listeria spp. and L. monocytogenes, surface water and point source pollution samples from an urban and a rural municipal water supply watershed in Nova Scotia, Canada, were examined over 18 months. Presumptive Listeria spp. were cultured from 72 and 35% of rural and urban water samples, respectively, with 24% of the positive samples containing two or three different Listeria spp. The L. innocua (56%) and L. welshimeri (43%) groups were predominant in the rural and urban watersheds, respectively. Analysis by the TaqMan assay showed a significantly (P < 0.05) higher prevalence of L. monocytogenes of 62% versus 17% by the culture-based method. Both methods revealed higher prevalences in the rural watershed and during the fall and winter seasons. Elevated Escherichia coli (>100 CFU/100 ml) levels were not associated with the pathogen regardless of the detection method. Isolation of Listeria spp. were associated with 70 times higher odds of isolating L. monocytogenes (odds ratio ‫؍‬ 70; P < 0.001). Serogroup IIa was predominant (67.7%) among the 285 L. monocytogenes isolates, followed by IVb (16.1%), IIb (15.8%), and IIc (0.4%). L. monocytogenes was detected in cow feces and raw sewage but not in septic tank samples. Pulsotyping of representative water (n ‫؍‬ 54) and local human (n ‫؍‬ 19) isolates suggested genetic similarities among some environmental and human L. monocytogenes isolates. In conclusion, temperate surface waters contain a diverse Listeria species population and could be a potential reservoir for L. monocytogenes, especially in rural agricultural watersheds. Listeria monocytogenes is a Gram-positive bacterium that is able to tolerate high salt concentrations, cold temperatures, and acid stress (1). Pathogenic Listeria species include L. monocytogenes and L. ivanovii (2), while 13 nonpathogenic species exist: L. cornellensis, L. grandensis, and L. riparia (3, 4).Listeria spp. are considered ubiquitous in the natural environment, and representatives of the genus, including L. monocytogenes, have been isolated from soil, surface waters, animal feeds, animal feces, sewage, food processing plants, and farm environments (5-11). In one New York study, L. seeligeri and L. welshimeri were dominant species in water and other environmental samples from pristine environments, while L. innocua and L. monocytogenes were associated with urban environments (8). Focusing on Listeria spp. diversity, Chapin et al. (12) reported finding Listeria spp. (excluding L. monocytogenes) in 22 and 51% of the surface water samples from pristine and produce growing areas, respectively, in New York. Linke et al. During a 1-year longitudinal study, water, sediment and water plants from two creeks and one pond were sampled monthly and analyzed for the presence of Listeria species. A total of 90 % of 30 sediment samples, 84 % of 31 water plant samples and 67 % of 36 water samples were tested positive. Generally, most probable number counts ranged between 1 and 40 g were detected. Species differentiation based on FT-IR spectroscopy and multiplex PCR of a total of 1220 isolates revealed L. innocua (46 %), L. seeligeri (27 %), L. monocytogenes (25 %) and L. ivanovii (2 %). Titers and species compositions were similar during all seasons. While the species distributions in sediments and associated Ranunculus fluitans plants appeared to be similar in both creeks, RAPD typing did not provide conclusive evidence that the populations of these environments were connected. It is concluded that (i) the fresh-water sediments and water plants are year-round populated by Listeria, (ii) no clear preference for growth in habitats as different as sediments and water plants was found and (iii) the RAPD-based intraspecific biodiversity is high compared to the low population density.Keywords: biodiversity; fresh water; Listeria monocytogenes; Listeria spp.; sediment; water plants.Dedication: This work is dedicated to the memory of Professor Peter Böger. -Microbial quality of baked egg products was evaluated by counting the levels of sanitary indicative bacteria (aerobic plate counts, coliforms, and E. coli), L. monocytogenes and Salmonella spp. at the critical control points (CCPs) of manufacturing process. In addition, the survival and growth of foodborne pathogens in various egg products (cheese, tuna, tteokgalbi, pizza omelets, baked egg, and steamed egg) were investigated at 4, 10, and 15 o C. The contamination level of aerobic plate counts decreased from 4.67 log CFU/g at CCP 1 to 0.56 log CFU/g at CCP 3 in baked egg products. No coliforms and E. coli were detected at all CCPs. Although L. innocua and Salmonella spp. were identified at CCP 1, no L. monocytogenes and Salmonella spp. were detected in the final products. The contamination levels of aerobic plate counts and coliforms in egg strips and number of aerobic plate counts in Tteokgalbi omelet are higher than the microbiological standard of processed egg products. At 10 o C, the growth of all pathogens was not prevented in omelet and baked egg, but the populations of S. Typhimurium and E. coli were reduced in steamed egg at 10 o C, regardless of the presence of other pathogens. The growth of L. monocytogenes was faster than that of S. Typhimurium and E. coli in omelet. More rapid growth of S. Enteritidis than S. Typhimurium was observed in egg products, indicating the greater risk of S. Enteritidis than S. Typhimurium in egg products.
