Lignin-Degrading Abilities of Novel Autochthonous Fungal Isolates Trametes hirsuta F13 and Stereum gausapatum F28

Abstract: SUMMARYThe aim of this research is to isolate and identify fungi with high lignin-degrading abilities that are autochthonous to southern Serbian region. Two novel fungal isolates identified as Trametes hirsuta F13 and Stereum gausapatum F28 were selected to assess their ligninolytic enzyme activities and the efficiency of lignin removal from beech wood sawdust. Obtained results show that both isolates are good sources of industrially valuable enzymes with a potential for application in various biotechnological… Show more

“…It is a basidiomycetous genus and some of its species were obtained as endophytes of coastal grasses and forest trees Theobroma gileri (Márquez et al 2008;Thomas et al 2008). Lastly, S. gausapatum collected from oak forest and orchards in southern Serbia displayed high lignin-degrading abilities and the efficiency of lignin removal from beech wood sawdust (Jović et al 2018). Not much information was reported on the plant growth promoting abilities of this strain.…”

“…The three isolates with the best lignocellulolytic enzyme activities were identified using ITS sequences, and the sequences were deposited in the NCBI GenBank database. Their accession numbers are KY264754.1 (Trametes hirsuta F13), KY264753.1 (Stereum gausapatum F28), and MF521930.1 (Myrmaecium fulvopruinatum F28) (Jović et al, 2018). Sugar beet MS was obtained from a local alcohol industry, and beechwood sawdust was obtained from a local sawmill.…”

“…Molecular identi cation of fungal isolates DNA extraction and PCR ampli cation were carried out according to the procedure described by Jović and coworkers with some modi cations [14]. For PCR ampli cation of the internal transcribed spacer (ITS) region of the ribosomal RNA coding DNA, a primer set ITS1/ITS4 was used.…”

“…The PCR ampli cation was performed in QB24 Thermal Cycler (Quanta Biotech Ltd., By eet, UK). The temperature parameters were set as follows: 15 min at 95 °C forinitial denaturation, 30 cycles of denaturation step (30 s at 95°C), annealing step (30 s at 55 °C) and elongation step (90 s at72 °C), and 5 min at 72 °C for nal extension [14].…”
