Ligand-Dependent Recruitment of the ErbB4 Signaling Complex into Neuronal Lipid Rafts

Abstract: Neuregulin (NRG) regulates synapse formation and synaptic plasticity, but little is known about the regulation of NRG signaling at synapses. Here we show that the NRG receptor ErbB4 was localized in anatomically defined postsynaptic densities in the brain. In cultured cortical neurons, ErbB4 was recruited to the neuronal lipid raft fraction after stimulation by NRG. Along with ErbB4, adaptor proteins Grb2 and Shc were translocated to lipid rafts by NRG stimulation. In transfected human embryonic kidney 293 cel… Show more

“…ErbB4, a member of the RTK family that binds neuregulin (NRG), also localizes to lipid rafts in a ligand-dependent manner in primary cortical neuronal cultures and human embryonic kidney (HEK293) cells (Ma et al 2003). Moreover, disruption of lipid rafts inhibits NRG-induced activation of ERK and prevents NRG-induced blockade of long-term potentiation at synapses in hippocampal slices (Ma et al 2003).…”

“…In the CNS, neuregulin stimulates recruit-ment of ErbB4 and Grb2 to neuronal lipid rafts. 203 Grb2 involvement in neuronal signalling is thus directly modulated by neuregulin, and DISC1 function may turn out to be similarly influenced by neuregulin and vice versa through its interaction with Grb2.…”

“…For example, lipid rafts are necessary for signaling pathways initiated by IgE (Baird et al, 1999), T-cell antigen receptor activation (Janes et al, 2001), and glial cell line-derived neurotrophic factor receptor activation (Tansey et al, 2001). In the nerve system, disruption of lipid rafts prevents neurons from responding properly to trophic factors, including neuregulin and BDNF (Ma et al, 2003;Suzuki et al, 2004).…”
