Molecular sequence analyses have yielded many important insights into diatom evolution, but there have been few attempts to relate these to the extensive fossil record of diatoms, probably because of unfamiliarity with the data available, which are scattered widely through the geological literature. We review the main features of molecular phylogenies and concentrate on the correspondence between these and the fossil record; we also review the evolution of major morphological, cytological and life cycle characteristics, and possible diatom origins. The first physical remains of diatoms are from the Jurassic, and well-preserved, diverse floras are available from the Lower Cretaceous. Though these are unequivocally identifiable as centric diatoms, none except a possible Stephanopyxis can be unequivocally linked to lineages of extant diatoms, although it is almost certain that members of the Coscinodiscophyceae (radial centrics) and Mediophyceae (polar centrics) were present; some display curious morphological features that hint at an unorthodox cell division mechanism and life cycle. It seems most likely that the earliest diatoms were marine, but recently discovered fossil deposits hint that episodes of terrestrial colonization may have occurred in the Mesozoic, though the main invasion of freshwaters appears to have been delayed until the Cenozoic. By the Upper Cretaceous, many lineages are present that can be convincingly related to extant diatom taxa. Pennate diatoms appear in the late Cretaceous and raphid diatoms in the Palaeocene, though molecular phylogenies imply that raphid diatoms did in fact evolve considerably earlier. Recent evidence shows that diatoms are substantially underclassified at the species level, with many semicryptic or cryptic species to be recognized; however, there is little prospect of being able to discriminate between such taxa in fossil material.

Many diatom and other microbial eukaryote morphospecies consist of a variable number of (pseudo)cryptic species, with obvious consequences for such fields as biogeography and community ecology. Here, we investigated the species limits of morphologically similar small-celled strains of the model diatom Sellaphora from the United Kingdom and Australia, using cox1 mitochondrial and rbcL chloroplast gene sequences. Based on cox1 sequence data, the sequenced strains belonged to six closely related lineages, presumably species, of which one corresponds to the previously described S. auldreekie D.g. mann & s.m. mCDonalD. Although rbcL displayed less sequence variation, the same six lineages were also recovered in an rbcL phylogeny of the genus. Molecular species discovery was compared to mating trials involving three of the lineages, showing that they were reproductively isolated. Incomplete evidence from a fourth lineage suggested that it too was reproductively isolated. A posteriori examination of light microscope morphology revealed no simple metrics or presence/absence characters that could consistently separate all species of the auldreekie complex, even though some do differ in pole width or stria density. While it is premature to make conclusions about their biogeography, it is obvious that a number of cryptic Sellaphora species thus far undetected in the UK are easily found at several localities in warm-temperate Australia.

A new species of raphid pennate diatom producing a blue pigment, Haslea karadagensis Davidovich, Gastineau & Mouget, sp. nov., was recently isolated from the Crimean coast of the Black Sea (Ukraine). This organism is very similar to the well-known Haslea ostrearia, the first described 'blue' diatom, which produces marennine, the pigment involved in the greening of oysters. The Ukrainian diatom, H. karadagensis, differs slightly from H. ostrearia in the structure of its frustule, and the two organisms are unable to interbreed. Two molecular markers, rbcL and the ITS1-5.8S-ITS2 sequences, showed 2% and >50% differences, respectively, between the two species. UV-visible spectrophotometry and in vivo confocal micro-Raman spectroscopy were used to compare the pigment of H. karadagensis with marennine. Both pigments showed absorption bands in the UV and red regions, but the positions of the maxima differ between the pigments. Significant differences were observed by microRaman spectroscopy in the 1000-1700 cm À1 wavenumber range, revealing that the pigments are different molecules. Haslea karadagensis is the first example of a new 'blue' diatom and produces a novel blue pigment.
