Levothyroxine: Conventional and Novel Drug Delivery Formulations

Abstract: Despite the fact that levothyroxine is one of the most prescribed medications in the world, its bioavailability has been reported to be impaired by many factors, including interfering drugs or foods and concomitant diseases, and persistent hypothyroidism with a high dose of levothyroxine is thus elicited. Persistent hypothyroidism can also be induced by noninterchangeability between formulations and poor compliance. To address these issues, some strategies have been developed. Novel formulations (liquid soluti… Show more

“…The American Thyroid Association recommends an intravenous levothyroxine dose of 75% of the oral dose. Intravenous injection is firmly not recommended for long-term treatment due to its rapid and significant effect on thyroid hormone homeostasis [ 10 ]. Hence, intravenous treatment was pursued for three days only.…”

“…T3, a thyroid hormone, governs a myriad of physiological processes encompassing growth, metabolism, body temperature, and heart rate [11]. With swifter and more potent physiological effects compared to thyroxine (T4), approximately 20% of T3 is directly secreted and synthesized by the thyroid gland, while the remaining 80% is converted from T4 in circulation [12]. T3's synthesis and secretion are stimulated by the thyroid-stimulating hormone (TSH) secreted by the pituitary gland, rendering it more biologically active than T4, despite its lower serum levels [13].…”
