“…In the last two decades, several studies have extensively reported on the presence of anti-hsp antibodies in various immune-mediated diseases (Tishler and Shoenfeld, 1996) (Panchapakesan et al, 1992), but such findings have been debated by others (Horvath et al, 2001, Gruber et al, 1996. Thus, Gruber et al (Gruber et al, 1996) found comparable levels of anti-hsp65 between patients with SLE and other controls,…”

“…At the antigen level, several Hp antigens have been considered important for the loss of immunological tolerance to myelin antigens, particularly heat shock proteins (hsp) (Chiba et al, 2006, Panchapakesan et al, 1992, Salvetti et al, 1992, Scotti et al, 2010, Tishler and Shoenfeld, 1996. Several approaches have been used to assess whether hsps are implicated in the pathogenesis of MS, including studies on immune responses to hsp in MS and the extent of cross-reactivity between hsp and CNS myelin, as well as the expression of hsps in the brain of patients with MS (Salvetti et al, 1992, Tishler and Shoenfeld, 1996, Horvath et al, 2001, Gruber et al, 1996, Chiba et al, 2006. Because of the conserved nature of hsp there is a high degree of A C C E P T E D M A N U S C R I P T ACCEPTED MANUSCRIPT 5 amino acid homology amongst hsp60 of different bacteria; thus, immune responses against hsps show a high degree of cross-reactivity (Van Eden, et.al.…”

“…Conroy et al (1996) found that there was a similar increase of antiHsp70 and anti-Hsp90 antibodies in children with SLE as in adult patients and suggested that SLE in children and adults might share a similar pathological mechanism of disease. Horvath et al (2001) investigated the presence of antibodies against both human Hsp60 and mycobacterial Hsp65 in patients with systemic lupus erythematosus, systemic sclerosis, undifferentiated connective tissue diseases (UCTD), primary Raynaud syndrome, rheumatoid arthritis, polymyositis; they found that the levels of anti-Hsp60 and anti-Hsp65 antibodies were significantly higher only in the UCTD patients. Shinghai et al (1995) reported the presence of antibodies to Hsps in pa- tients with autoimmune liver diseases and suggested that the presence of anti-Hsc70 antibodies was an indicator for the disease activity of primary biliary cirrhosis.…”

“…However, depending on the Ag used in the assays and the method of detection, titer and frequency of anti-HSP60 autoAb vary from one study to another. Whereas serum concentration of anti-HSP60 did not differ from controls, in an ELISA based on the recognition of 0.1 µg recombinant HSP60 [30], the IgG titer was higher in SLE patient than in controls, in an ELISA based on the use of 50 ng human mitochondrial HSP60 as a captured Ag [31]. Anti-HSP60 autoAb was found in 26% of SLE patients and controls, with 500 ng/ml recombinant HSP60 as coated Ag [32]; there were 39% of SLE patients, compared with 4% of healthy controls with 0.5 µg recombinant HSP60 [6].…”
