Leukemogenic Activity of Filtrates from Radiation-Induced Lymphoid Tumors of Mice

Abstract: Cell-free filtrates of x-ray-induced lymphoid tumors of strain C57BL/ Ka mice have elicited, on injection into newborn isologous hosts, a lymphoma incidence of 15 to 19 percent. In control mice of the same subline, the incidence of spontaneous lymphoma is about 1 percent. No leukemogenic activity could be detected in filtrates from thymi harvested at 2 to 32 days following completion of x-ray treatment. Activity was evident at 64 days and was perhaps somewhat greater at 128 days. Serial cell-free passage of fi… Show more

“…The successful isolation of a filterable MIuLV' from chemically iinduiced lvympho- mata (Haren-Ghera, 1967;Toth, 1963;Irino, Ota and Sezaki, 1963;Ball and McCarter, 1971) and radiation induced lymphomata (Leiberman and Kaplan, 1959;Kaplan, 1967;Latarjet and Duplan, 1962) has been reported before. MuLV from oestrogen induced lymphomata has been successfully isolated for the first time by Kunii, Takemoto and Furth (1965 (Lilly, 1970;McDevitt and Bodmer, 1972).…”

“…C57BL/Ka is a substrain of strain C57BL (H-2 b) maintained by Dr. Henry S. Kaplan at Stanford University. This substrain is characterized by 90-100% lymphoma incidence after whole body irradiation (9,10). Briefly, thymus, spleen, and lymph nodes were removed, pooled, homogenized in phosphate-buffered saline (PBS) to make a 20% suspension (vol/vol), and centrifuged at 10,000 rpm for 15 rain.…”

“…RadLV preparations were cell-free extracts obtained from virus-induced lymphoid tumors of C57BL/Ka mice as previously described (9,10). C57BL/Ka is a substrain of strain C57BL (H-2 b) maintained by Dr. Henry S. Kaplan at Stanford University.…”
