Lessons from Drosophila Pointed, an ETS family transcription factor and key nuclear effector of the RTK signaling pathway

Abstract: Summary The ETS family of transcription factors are evolutionarily conserved throughout the metazoan lineage and are critical for regulating cellular processes such as proliferation, differentiation, apoptosis, angiogenesis, and migration. All members have an ETS DNA binding domain, while a subset also has a protein–protein interaction domain called the SAM domain. Pointed (Pnt), an ETS transcriptional activator functions downstream of the receptor tyrosine kinase (RTK) signaling pathway to regulate diverse pr… Show more

“…Rlb was localized in a small region of chromosome 2R (Hita et al., 1999, 2006) that contains two genes: CG33136 and edl / mae (ETS-domain lacking)/(modulator of the activity of ETS). Interestingly, Edl interacts via its SAM (sterile alpha motif) domain with several ETS transcription factors (Baker et al., 2001) such as the transcriptional activator Pointed (mainly Pointed P2) and the repressor Yan/Aop (Vivekanand, 2018), both involved in the EGF pathway. When activated, the EGF signal triggers the mitogen-activated protein kinase/extracellular signal-regulated kinase (ERK) kinase (MEK) known as Rolled.…”

“…When activated, the EGF signal triggers the mitogen-activated protein kinase/extracellular signal-regulated kinase (ERK) kinase (MEK) known as Rolled. Among many targets, rolled phosphorylates Pointed and Yan, thus modulating their function by changing their SAM domain binding affinity (Vivekanand, 2018). The universal output of the EGF pathway is induction of gene expression, mostly through the Pointed transcriptional activator.…”

“…As a first example, we investigate the ETS family transcription factor Pointed (Pnt), a known regulator of differentiation and proliferation in multiple fly tissues with implications in hemocyte development (Zettervall et al 2004; Dragojlovic-Munther and Martinez-Agosto 2013; Shwartz et al 2013; reviewed in Vivekanand 2018). Pnt transcript is expressed at very low levels in the PSC, MZ1-1, MZ2-1, and MZ2-2 (Figure 4A-B) and its level rises significantly in MZ2-3 (Figure 4B).…”

“…EGFR signals via the canonical Ras/MAPK to activate the transcription of target genes in a context-dependent manner via the Pointed ETS transcription factor [61]. Its expression during development is broad and therefore is not considered to be the critical aspect regulating its activity [62].…”
