Less Is More: Ebola Virus Surface Glycoprotein Expression Levels Regulate Virus Production and Infectivity

Abstract: The Ebola virus (EBOV) surface glycoprotein (GP 1,2 ) mediates host cell attachment and fusion and is the primary target for host neutralizing antibodies. Expression of GP 1,2 at high levels disrupts normal cell physiology, and EBOV uses an RNA-editing mechanism to regulate expression of the GP gene. In this study, we demonstrate that high levels of GP 1,2 expression impair production and release of EBOV virus-like particles (VLPs) as well as infectivity of GP 1,2 -pseudotyped viruses. We further show that thi… Show more

“…Transfections were performed as previously described (Mohan et al., 2015, Urbanowicz et al., 2016). Briefly, 1.2x10 6 HEK293T cells were seeded overnight in a 10cm diameter Primaria coated dish (Corning) in 10mL of DMEM supplemented with non-essential amino acids and heat-inactivated FBS.…”

“…Within endo/lysosomal compartments, host endosomal cysteine proteases (cathepsins) cleave the filoviral GP1 protein to generate an entry intermediate comprising an N-terminal GP1 fragment and GP2 (12)(13)(14). Underlying the viral mem-brane is a viral matrix comprised mainly of viral protein 40 (VP40).…”
