Lentiviral Vector-Mediated shRNA against AIMP2-DX2 Suppresses Lung Cancer Cell Growth through Blocking Glucose Uptake

Abstract: Aminoacyl-tRNA synthetases [ARS]-interacting multifunctional protein 2 (AIMP2) has been implicated in the control of cell fate and lung cell differentiation. A variant of AIMP2 lacking exon 2 (AIMP2-DX2) is expressed in different cancer cells. We previously studied the expression level of AIMP2-DX2 in several lung cell lines and reported elevated expression levels of AIMP2-DX2 in NCI-H460 and NCI-H520. Here, we report that the suppression of AIMP2-DX2 by lentivirus mediated short hairpin (sh)RNA (sh-DX2) decre… Show more

“…Note, knowledge on the role of aminoacyl-tRNA synthetases in cancer is just emerging [76] and through the use of a lentiviral mediated shRNA vector, a link between aminoacyl-tRNA synthetases [AARS]-interacting multifunctional protein 2 (AIMP2) and repressed EGFR signalling was established that resulted in repressed glucose uptake [77]. We also observed induced expression of heterogeneous ribonucleoprotein (hnRNP) that takes on diverse functions in the processing of mRNA.…”

“…For example, suppression of AIMP2-DX2 using lentivirus-mediated shRNA in lung cancer cells inhibited cell growth by blocking glucose uptake and growth factor mediated signaling pathways [152]. A trans-splicing ribozyme was used to induce selective death of AIMP2-DX2 expressing lung cancer cells [153].…”

“…AIMP2-DX2 is closely correlated to the severity of lung cancer and is positively related to drug resistance in ovarian cancer [ 16 , 17 ]. Inhibition of AIMP2-DX2 may suppress the proliferation of lung cancer cells and restrain the tumor formation in nude mice [ 18 , 19 ]. Thus AIMP2-DX2 acts as an oncogene, different from AIMP2.…”
