Leishmanicidal Evaluation of Novel Synthetic Chromenes

Abstract: In the present paper, twelve chromenes were synthesized by coupling of 2,2,8,8-tetramethyl-8H-pyrano[2,3-f]chroman-4-one 1 with various aryl and benzylmagnesium chlorides. The synthetic compounds were examined for in-vitro activity against Leishmania major, and some of them displayed efficient anti-leishmanial activity. Among the compounds tested, compounds 9 (4-(2-chloro-benzylidene)-2,2,8,8-tetramethyl-3,4-dihydro-2H,8H-pyrano[2,3-f]chromene 9a and 4-(2-chloro-benzyl)-2,2,8,8-tetramethyl-2H,8H-pyrano[2,3-f]c… Show more

“…Chromenes have repeatedly been reported to possess antiprotozoal activity (Ganapaty et al, 2006;Alizadeh et al, 2008b;Batista et al, 2008). In order to investigate the potential role of encecalol angelate as an antiprotozoal principle in A. conyzoides, the synthetic product was tested in vitro against the four protozoan parasites, i.e.…”

“…1 This study is part of the doctoral thesis of Dipak Harel (University of Münster). antiprotozoal activity (Ganapaty et al, 2006;Alizadeh et al, 2008b;Batista et al, 2008). During our continuous search for plant constituents with antiprotozoal activity, we recently discovered that the dichloromethane extract of Ageratum conyzoides L., a medicinal plant widespread in tropical and subtropical regions and used in folk medicine of many countries for a wide variety of indications including sleeping sickness (Johnson, 1999;Kamboj and Saluja, 2008), displayed considerable in vitro activity against Trypanosoma brucei rhodesiense, etiologic agent of East African Human Trypanosomiasis or Sleeping Sickness, with an IC 50 value of 0.78 g/mL (Nour et al, 2009(Nour et al, , 2010.…”

“…In this study, a chromene exhibited selective docking to Leishmania major N-myristoyltransferase, showing that chromenes may be promising as antiparasitic based on in-silico analysis. In other study, it was evaluated the leishmanicidal activity of twelve synthetic chromenes against in vitro L. major promastigotes, and some compounds exhibited important leishmanicidal effect (% growth inhibition>70%), showing the potential biological of this chemical class (Alizadeh et al, 2008).…”

“…Several studies have demonstrated the insecticidal, antibacterial, fungicidal and cytotoxic activities of these substances (Bandara et al, 1992;Burkhardt et al, 1994;Iqbal et al, 2004;Chen et al, 2005). Furthermore, some compounds of this class of natural products have been described to have notable antiprotozoal effect (Alizadeh et al, 2008;Batista Jr. et al, 2008;Harel et al, 2011).…”

“…Chromene-based compounds have been reported to possess many pharmacological activities, including antibacterial properties [6,7]; however, recent reports demonstrated the potential of 4-aryl-4H-chromenes 1-3 as apoptosis inducers (Fig. 1).…”
