Leishmania spp Epitopes in Humans Naturally Resistant to the Disease: Working Toward a Synthetic Vaccine

Abstract: Vaccines are one of the most effective strategies to fight infectious diseases. Reverse vaccinology strategies provide tools to perform in silico screening and a rational selection of potential candidates on a large scale before reaching in vitro and in vivo evaluations. Leishmania infection in humans produces clinical symptoms in some individuals, while another part of the population is naturally resistant (asymptomatic course) to the disease, and therefore their immune response controls parasite replication.… Show more

“…Then, epitopes predicted by immunoinformatics are submitted to further experiments in vitro and in vivo for confirmation. Leishmania antigenic proteins validated by experiments usually are screened for their epitopes of T and B cells by immunoinformatics ( 2 , 30 , 47 , 48 ). However, this approach is mainly used to construct multi-epitope vaccines against VL and relies on known antigenic proteins.…”

“…Then, a developer solution was added, and plates were submitted for reading on a Flow cytometer FACSCanto II (Becton Dickinson BD Biosciences, San Diego, CA, USA). Results were expressed as the Stimulation Index (SI), calculated by dividing the results of the stimulated number of cells by each peptidomimetic antigen regarding the number of cells without stimulation and data were processed with Graph Pad Prism 8.0.2 software according to protocols published elsewhere [ 52 ].…”

“…Antimicrobial peptide-based antileishmanials, vaccines, and cytokines 10,14 have been reported against leishmanial parasites as they offer specific immune responses. 15,16 Certain approaches such as peptide vaccines, 17 stapled peptides, 16 and nanoparticle-based drug delivery 18 for induction of rapid immune response have been utilized. 19,20 Polymer poly( d , l -lactide- co -glycolide) (PLGA) nanoparticles (NPs), functionalized with a TNFα-mimicking octapeptide have also been shown to confer protection against VL.…”
