Lefty Contributes to the Remodeling of Extracellular Matrix by Inhibition of Connective Tissue Growth Factor and Collagen mRNA Expression and Increased Proteolytic Activity in a Fibrosarcoma Model

Abstract: Homeostasis of the extracellular matrix (ECM) of tissues is regulated by controlling deposition and degradation of ECM proteins. The breakdown of ECM is essential in blastocyst implantation and embryonic development, tissue morphogenesis, menstrual shedding, bone formation, tissue resorption after delivery, and tumor growth and invasion. TGF-␤ family members are one of the classes of proteins that actively participate in the homeostasis of ECM. Here, we report on the effect of lefty, a novel member of the TGF-… Show more

“…In a fibroblastic tumor model in vivo (58,59), it was shown that lefty decreased collagen type I mRNA expression and simultaneously increased collagenolytic, gelatinolytic, elastolytic, and caseinolytic activities, similar changes as seen in the current studies with Smad3 KO mice. Moreover, in a recent study, Morris demonstrated that the loss of ␣ v ␤ 6 integrin in mice induces a MMP-12-dependent emphysema, related to the lack of activation of latent TGF-␤ (10).…”

“…To test enzymatic activity, equal mole ratios of GST/TSP50, GST/TSP50/T/A, and GST proteins were applied in duplicate to 12% Tris-glycine gels containing h-casein or gelatin (Bethesda Research Laboratory, Bethesda, MD; refs. 26,27). The positive control for the h-casein gel was homogenous trypsin type IX (Sigma), whereas collagenase type VII (Sigma) was used for the gelatin gel.…”

“…Normal testis and breast tissue RNAs were purchased from Clontech and served as positive and negative controls, respectively. To evaluate the TSP50 gene expression levels in different cell lines, semiquantitative reverse transcription-PCR (RT-PCR) was done using the Titan One tube RT-PCR system (Roche) as previously described (26,29). The sense and antisense primers for TSP50 and GAPDH (internal control) were also previously described (1,22,29).…”

“…LEFTY1 (also known as LEFTY-B), another promising target gene in the bone metastases profile, was confirmed to be significantly upregulated in bone metastasis. LEFTY1 was reported to be a secreted molecule of the TGF-β superfamily involved in the Nodal signaling pathway, and a marker of the stemness of cells (31-33), and to contribute to the remodeling process of the extracellular matrix (34). In contrast, LEFTY1 also plays an important role as an inhibitor of Nodal, a crucial component involved in metastatic melanoma cells, in a negative feedback mechanism (reviewed in ref.…”
