Lectins in Castor Bean Seedlings

Abstract: The amounts of the two lectins (ricin and Ricinus communis agglutinin) in tissues of castor bean seedlings were followed during germination and early growth. For measurement, lectins in extracts were separately eluted from Sepharose columns; an antibody to the agglutinin was also used to detect the lectins by immunodiffusion. The endosperm of the dry seed contains 3.5 mg total lectin (5.6% of the total seed protein), which declines by 50% by day 4 and more rapidly thereafter as the tissue is completely consume… Show more

“…The AB toxin ricin (Figure 1) is produced in the developing endosperm of the seeds of the castor oil plant, Ricinus communis (Harley and Beevers 1986), as a preproprotein that is matured by proteolytic cleavage. The mature heterodimeric holotoxin comprises a catalytically active A chain (RTA) disulphide-linked to a lectin B chain (RTB).…”

“…4,12 RCA 120 causes agglutination and lysis of mammalian red blood cells. 4,13 It is a tetramer consisting of 2 ricinlike heterodimers held together by noncovalent forces. 4 Each heterodimer consists of an A chain (32 kDa) linked to a galactose-binding B chain (37kDa), both of which are similar in structure to the A and B chains of RCA 60 .…”

“…While globulins comprise of storage proteins, albumins form a heterogeneous class of unrelated proteins and include metabolic, structural, and/or regulatory proteins (Vioque et al, 1999). Many albumins such as lipoxygenases (Clemente et al, 2000), glycosidases (Agrawal and Bahl, 1968), proteinases (Schlereth et al, 2001), proteinase inhibitors (Kochhar et al, 2000), and lectins (Harley and Beevers, 1986) have been demonstrated to exhibit distinct functions. Recently, another protein belonging to albumin class, referred to as albumin 2 from garden pea (Pisum sativum) has been demonstrated to have regulatory role in polyamine metabolism (Vigeolas et al, 2008).…”
