Lectins as Biomarkers of IC/BPS Disease: A Comparative Study of Glycosylation Patterns in Human Pathologic Urothelium and IC/BPS Experimental Models

Abstract: Pathophysiology of interstitial cystitis/bladder pain syndrome (IC/BPS) remains poorly understood, as well as its effective diagnosis and therapy. Studying changes in tissue glycosylation patterns under pathological conditions is a promising way of discovering novel biomarkers and therapeutic targets. The glycobiology of IC/BPS is largely understudied, therefore we compared glycosylation patterns of normal human urothelium with the urothelium of IC/BPS patients using a selection of 10 plant-based lectins with … Show more

“…Chronic inflammatory conditions can significantly affect the glycosylation profile of urothelial cells, contributing to impaired barrier function [3,35]. The importance of restoring a deficient urothelial glycocalyx is also reflected in the use of glycosaminoglycan replenishment as one of the most common and successful therapy options used for IC/BPS [48].…”

“…The glycosylation profile of urothelial cells is altered in a chronically inflamed bladder, and a loss of the protective GAG layer may contribute significantly to an increased permeability of the urothelium [3,35].…”

“…Furthermore, IC/BPS cell models could also originate from cancer cells. For example, human carcinoma epithelial RT4 cells and T24 cells were treated with TNF-α to mimic the inflammatory condition in IC/BPS [ 160 , 161 ]. Ketamine has been successfully used as a stimulus to induce dysregulation in SV-HCU-1, RT4, and 5637 cell lines [ 162 ].…”

“…Particularly, potassium ions are suspected to provoke nerve terminals of C-fibres triggering pain and OAB symptoms [12]. Using an animal model with cyclophosphamide (CYP) induced cystitis, Peskar et al [13 ▪ ] observed that changes in glycosylation patterns impairing the GAG-layer function may contribute to BPS/IC disease. The positive effect of restoring the GAG layer in patients with HIC or glomerulations using orally administrated pentosane polysulfate sodium could be demonstrated in a recently published systematic review and meta-analysis [14 ▪▪ ].…”
